Objective To study the activation of astrocytes (Astrocyte, Ast) expression of the epidermal growth factor receptor (epithelialgrowth factor receptor, EGFR) , and explore the EGFR played a key role in the activation of astrocytes . Shaking culture method combined with differential activation group affixed wall separation and purification of cultured astrocytes , the second generation of astrocytes will be divided into a control group , and the suppression group : routine containing 10? S DMEM medium 24h after astrocytes cells as the control group , with 20ng/ml ciliary neurotrophic factor , 10 ? astrocytes S in DMEM culture medium 24 h after activation group cells containing 30μmol / L effect of Genistein 10 ? S DMEM medium role in the activation group cells incubated for 24 h after cell as inhibitory cells . The observed changes in the morphology of the cells by immunofluorescence ; semi - quantitative RT-PCR analysis fibrillary acidic protein expression between cells ( glial fibrillary Irregularity in the protein , GFAP) in mRNA and EGFR mRNA expression . Activation group and the control group compared with the results the increase in the number of astrocytes cells , larger cell body processes increased variable length reticulation ; RT-PCR shows not only GFAP mRNA expression increased the , EGFR mMRA expression also significantly increased , with the control group were significantly different ( P <0.01 ) ; application EGFR inhibitor Genistein intervention , compared with the activation of the group , no longer increase the number of astrocytes smaller cell bodies , processes reduce shortened astrocyte activation suppression of GFAP mRNA expression decreased activation group were significantly different ( P <0.01 ) . Conclusion : 1 . Astrocytes activated EGFR expression is significantly upregulated by inhibiting EGFR expression ; 2.Genistein astrocyte activation can be inhibited .
