Abstract


Metal ions in a wide range of life processes in vivo , they maintain proteins, nucleic acids and other biological macromolecules of the basic structure, to achieve normal life functions of the body plays a very important role. Copper is the third in vivo levels of trace metals , the body for the intracellular concentration of copper ions have a very strict regulatory mechanism of copper excess or deficiency can lead to disease. The copper ions are often used pesticides fungicides for inhibiting the growth of bacteria and fungi , but improper use can cause injury and even prevention through bioaccumulation directly endanger human health or the environment and therefore the organism of copper ions detection has important applications, it has long been sought in living cells can be detected with high resolution copper ion distribution methods . In this thesis, a copper ion regulation from Escherichia coli transcriptional promoter CueR protein and yellow fluorescent protein fusion constructs can respond to copper ions fluorescent protein probes CuFP. By means of genetic engineering to further probe the gene deletion mutagenesis and obtained a series of mutants. We use a variety of metal ions were titrated and studied these CuFP fluorescent protein probes the changing nature of the final screening with copper ions that can respond to specific fluorescent protein probes and silver ions are able to respond to specific fluorescence protein probes. This thesis probes will be used to obtain intracellular copper ions and silver ion detection, or pesticide residues and heavy metal contamination is detected.
