Diversity of Achaea Communities in Surface Sediment Located at Yellow River Delta

Abstract


Objective : To study the diversity and distribution near the coast of the Yellow River Delta area archaea . Methods : PCR technology based build archaeal 16S rDNA gene library to choose from six different stations sediment samples to conduct research on a small scale . Results: 1 . Study treatment received a total of 568 clones , amplification of the 16S rRNA gene after restriction digestion analysis and application software DOTUR of a 97% cut off after a total of 73 operational taxonomic units (Operational Taxonomic Unit, OUTs). All positive clones were sequenced after double digestion by online through traditional statistical methods, and the phylogenetic tree was constructed by DNA sequence alignment . Of the the experimental 16S rDNA analysis found that the archaea the Quansheng archaea (Crenarchaeota) , and Guangsheng archaea (Euryarchaeota) are from two categories . Wherein , Marine Crenarchaeotic Group Ⅰ (MG - i ) ( 88.8% ) occupy a major position , followed by the of the Miscellaneous Crenarchaeotic Group (MCG) ; DSAG ( Deep- Sea Archaeal Group ) the called MBG - B ; , MBG -D , also known as MG-Ⅲ (Marine Group Ⅲ) and TMEG ( the Terrestrial Miscellaneous Eurarchaeotal Goup ) . Conclusion: The results reflect the number of clones of the two MG- I and MCG ancient lineages 16SrDNA 96% of the total number of clones , both of which are the the ocean most common ancient lineages , which is also in line with the offshore environment is more stable features. The offshore sediment Medieval bacterial community structure in a horizontal range does not significantly change , just as the widening of the distance of the opposite coasts , the diversity of ancient lineages correspondingly high . Other geographical analysis , the study of ancient bacteria homologous sequences from more sediments of the river basin , some archaea reflect the regional characteristics of the anaerobic oxidation of methane may exist in the region , and thus indirectly reveals the community structure of archaea features should have a greater association with the characteristics of the coastal waters and its surrounding environment , the distribution and diversity of its community structure can also serve as an important indication of the marine environment , in order to facilitate the follow-up study of the field of microbiology .
