Abstract


Günther (Cynoglossus semilaevis) is one of the major in marine fishes , has important economic value . The farming area is located in the north of China , especially the Shandong Peninsula . Its foundation immune science research but there is very little , to depth study half slip tongue sole of the immune defense mechanism , our use of homologous cloning of get leukocyte mediated Su 8 ( Interleukin 8 , IL-8 ) gene and analysis of its gene expression and biological activity. Interleukin-8 is involved in inflammation in a mammal chemokine , belongs to the CXC family , also referred to as CXCL8 . In this paper, homologous cloning Günther full-length sequence of the gene IL-8 (CsIL-8) , and were identified and analyzed . Csil - 8 consists of 100 amino acid residues, and fish lineages 1 on the homology of IL-8 closer . It contains the CXC chemokine domain , including four conserved cysteine ??residues , wherein the composition of the two cysteine ??obvious the CXC structure . Using real-time quantitative PCR analysis of the organization of the gene expression characteristics under normal physiological conditions found CsIL-8 expression in various tissues , the bacteria after stimulation and subunit vaccine CsIL - 8 expression raised . The purified recombinant protein was able to induce rCsIL-8 Günther ? Peripheral blood leukocytes and head kidney lymphocyte migration and stimulate the proliferation of these cells ; When rCsIL-8 joined the head kidney lymphocyte culture medium , with the prolonged incubation time , rCsIL -8 to varying degrees raised the expression of IL-1β , and IL-8 .
