For improving bioavailability of chicken transfer factor, extending action duration invivo, reducing the utilization and decreasing the cost of animal husbandry production, thestudy was establishied, and the sustained release properties of liposomes were also beconsidered to explore the preparing methods of chicken transfer factor liposome(CTFL)andresearching physical&chemical properties, biological characteristics and immune efficiencyof vaccine. It will provide a new pathway for preventing and treating disease of animalhusbandry instead of CTF in China in future.Chicken transfer factor solution was prepared by homogenate, freeze-thaw, dialysis andother technology from healthy chicken spleen. It was yellowish and slightly fishy odor withpH 6.7; non-bacteria, non-toxic, non-sensitivity non-pyrogen and non-protein. The averagevalue of OD260/OD280 was 2.44 and multi-peptide content was 1.15mg/mL. Seventeenamino acids were contained in the CTF solution, glutamine (21.162μg/mL) was the highestand methionine (2.635μg/mL) was the lowest. The total content of amino acids was122.728μg/mL. All of values above were accorded with TF quality standards.The chicken transfer factor liposome was prepared by modified reverse phaseevaporation method with soy lecithin, vitamin E, cholesterol and other material. The solutionwas translucent milky white, tasteless; pH was 6.5; Non-bacteria,non-toxic, non-pyrogen andnon-allergic were appeared by different tests, it’s the same as CTF; The envelopingpercentage of CTFL was 56.50% at the day prepared, its leakage rate was 47.49% and pHwas 6.0 after conserving 6 months. The CTFL can be conserved as long as 6 months.For testing the immune efficiency of the chicken transfer factor prepared in thelaboratory, radix astragali, levamisole and ganoderma spore powder were taken ascomparison objects, and saline as control. The antibody titers of ND and EDS were detectedby HI test. After statistical analysis, the results showed that chicken transfer factor had a goodimmune efficiency on ND and EDS,and it is much better than others.For testing the influence of vaccine immune of the CTFL, the antibody titers of ND andEDS were detected by HI test with saline, liposome and CTF as control. The results ofstatistical analysis showed that there are significant differences between CTFL group and thecontrol groups, and immune efficiency of EDS is better than ND. These show that thechicken transfer factor liposome will be used widely in veterinary clinic and it will worthfuther exploitation.
