Transformation and Functional Analysis of a Novel Cold Responsive Glycosyltransferase-like Gene (OsCrGtl) in Rice

Abstract


OsCrGtl (Oryza sativa L. cold responsive glycosyltransferase-like gene) was a novel stress responsive gene identified in the indica cultivar Pei’ai 64S.It is highly induced by abiotic stresses,especially by cold, in both leaf and panicle at all the development stages analyzed in rice. It encoded a protein containing a glycosyltransferase domain, so it is a glycosyltransferase-like gene. In order to analyze its functions in cold tolerance or others, a binary vector pCAMBIA 1300-OsCrGtl,driven by strong constitutive promoter 2xCaMV 35S, was constructed, and OsCrGtl was then transformed into rice indica varieties 93-11 which is sensitive to cold through agrobacterium-mediated method. Nine independent transgenic rice plants were obtained. The cold tolerance of transgenic rice plants were assessed for their To and T1 generations seedlings. The results suggest that the transgenic rice seeds(T1) posses higher germinating potentiality than wild type ones under low temperature(15℃)although they shows no significant difference in their germinating rates. The electrolyte leakage (%) of the transgenic rice plant (To) increases slower and its free proline content accumulates faster than those of the wild-type 93-11 after cold treatment in chamber. The results also showed that the electrolyte leakage of the T1 generation transgenic seedlings grows slower, its soluble sugar and free proline content accumulate faster, and the activity of its POD. one of ROS-scavenging enzymes, is higher than those of the wild-type 93-11 after cold treatment. All these results suggest that the transgenic rice 93-11 plants were more tolerant to cold stress than wild-type ones. Additionally, the agricultural characters of To generation plants were investigated compared with wild type. The results suggest that the transgenic rice performed as well as wild-type under the normal condition.
