Abstract


Objective:Observe the degradation of nucleic acid in Schwann cell of sciatic nerve. Analysis the relationship between PMI and the degradation of nucleic acid in Schwann cell of sciatic nerve.Methods: the 27 SD rats were randomly divided into 9 groups with different time periods: 0h、6h、. 12h、18、24h、36h、48h、60h、72h. so we cut homologous sciatic nerves in different time periods. The sciatic nerves were dyed with methyl green pyronin staining.Results: longitudinal section of sciatic nerve: the nucleus of Schwann cell is stained in blue-green. The morphological structure of nucleus is intact、full and spindly. The cytoplasm of Schwann cell is stained with pink. The cell can be observed on anywhere. The number of cells is about 20-30. the cross section of sciatic nerve is less easily observed and studied.6-12h: the change of the staining of nucleus and the morphological structure is slight. 18-48h: the number of Schwann cells is down significantly: 10-20 per high power field. The nuclear staining tinged at 18h, a reduction in the size of 48h. 60-72h: the number of Schwann cells is about 8-10 per high power field. In the other regions, the staining tinged and structure become blur.the liner regression equation of average aimed area in sciatic nerve: Y=98.046-5.265X, R~2=0.82, P<0.01; the liner regression equation of average gray in sciatic nerve: Y=203.270-5.873X, R~2=0.711, P<0.01; the liner regression equation of Average Optical Density in sciatic nerve: Y=0.203-0.010X, R~2=0.509, P<0.01..Conclusion: there is a liner correlation between PMI and the degradation regularity of nucleic acid of Schwann cell in sciatic nerve. the liner regression equation of average aimed area in sciatic nerve is the best one: Y=98.046-5.265X, R~2=0.82, P<0.01.
