Abstract


OBJECTIVE:By measuring the expression level of GP73 in human hepatocellular carcinoma tissue, pericancerous tissue and normal liver tissue, the expression level was compared in these tissues, the relationship between the expression level and the clinicopathological features was analyzed.MATERIALS AND METHODS:Real-time fluorescence quantitative PCR (Real-time Q-PCR, SYBR Green method) was used to measure the expression level of GP73 in HCC tissue in 22 patients with HCC without preoperative chemotherapy and radiotherapy, pericancerous tissue in 11 patients with HCC without preoperative chemotherapy and radiotherapy and normal liver tissue in 17 patients without hepatitis and HCC. GAPDH was used as endogenous control. The relationship between the expression level of GP73 and the clinicopathological features was analyzed by SPSS version 17.0 statistical software.α=0.05 was regarded as the statistical criterion.RESUITS:1. The expression level of GP73 in HCC tissue was significantly higher than that in normal liver tissue (P<0.01).2. The expression level of GP73 in HCC tissue was significantly higher than that in pericancerous tissue (P<0.05). 3. The expression level of GP73 in pericancerous tissue was slightly higher than that in normal liver tissue (P> 0.05).4. The expression level of GP73 in HCC tissue was significantly correlated with HBV, cirrhosis, tumor diameter, daughter nude and tumor emboli (P<0.05); but it was not correlated with AFP and histological grade (P>0.05).CONCIUSION:1. The expression level of GP73 was significantly up-regulated in HCC, and it was correlated with HBV, cirrhosis, tumor diameter, daughter nude and tumor emboli, it may play an important role in the carcinogenesis of HCC.
