Expression of Synaptotagmin I and Effect of Thyroxine Treatment in the Prefrontal Lobe of Adult-onset Hypothyroid Rats

Abstract


ObjectiveDrugs was used to establish adult-onset hypothyroid rats and treatment models.To observe the expression of synaptotagmin I（Syt I） protein in frontal lobe of adult-onset hypothyroidism rats and the effects of different dosage therapy by levothyroxine（L-T4） on syt I protein.Further to explore the molecular mechanism of brain impairment by hypothyroidism.MethodsAdult male Sprague-Dawley rats weighing 250300g were used in current experiments. Fouty-four rats were divided into 4 groups randomly, The hypothyroid rats and treatment models were induced by drugs. （1） Hypothyroid group. Eleven hypothyroid rats were induced by daily intraperitoneal injection of 6-n-propyl-2-thiouracil （PTU） （dissolved in saline solution, 1 mg/100g body weight （BW）） for six weeks; （2） T4-5 group. Eleven rats were treated with PTU for six weeks as described above. But from the fifth week, they were also treated with intraperitoneally injected L-T4 （dissolved in saline solution, 5μg/100g BW） every day for two weeks; （3） T4-20 group. Eleven rats were treated according to the same protocols as the T4-5 group except increasing the dosage of L-T4 to 20μg/100g BW. （4） Control group. Eleven control rats were given same volume of saline solution for six weeks with hypothyroid rats. The radioimmunoassay kits were applied to assay the levels of serum T3 and T4, and immunohistochemical method was used to determine the levels SytⅠin the prefrontal lobe of rats from different groups. All statistical analysis was performed using SPSS software （version 16.0）. Values are expressed as mean±S.E.M. The data among the groups were compared using one-way ANOVA. Statistical significance was defined as P < 0.05.ResultsIn the hypothyroidism group, the levels of serum T3 and T4 were significantly lower than those in the control group（P< 0.05 or P< 0.01）, the levels of SytⅠprotein in the molecular layer（Ⅰlayer）、external granular layer（Ⅱlayer）、external pyramidal layer（Ⅲlayer）、internal granular layer（Ⅳlayer）、internal pyramidal layer（Ⅴlayer） of prefrontal lobe were significantly lower compared to the control group（P < 0.05 or P < 0.01）.In the T4-5 group, the serum T3、T4 levels of rats were similar to those in the control group（P > 0.05）, and showed signifcantly improvement of SytⅠprotein,which were similar to that in the control group（P > 0.05）;In T4-20 group, the levels of serum T3 and T4 were higher than the control group（P < 0.01）,the expression of syt I protein in theⅠ、Ⅱ、Ⅲ、Ⅳ、Ⅴlayers of the prefrontal lobe were similar to that in the control group（P > 0.05）. Conclusion These results suggest that adult-onset hypothyroidism caused down-regulation of Syt I level in theⅠ、Ⅱ、Ⅲ、Ⅳ、Ⅴlayers of the prefrontal lobe, which could be restored by L-T4 treatment, and routine dosage of L-T4 treatment could reverse the decrement of syt I protein.
