YiKang soup blastocyst implantation dysfunction improving effect and mechanism of action

Abstract


Objective To investigate the effect of YiKang decoction on the implantation dysfunction induced by mifepristone and it’s possible mechanism.Method All mice of 4-6 weeks were divided into four groups in random:normal group, model group, traditional Chinese medicine group (TCM group) and control group. Mice of TCM goup were further divided into low、moderate、high dosage group, and intragastric administration with 0.4、0.6、0.8ml YiKang decoction respectively for 28±7 days, while mice of control group were administration with 0.8ml saline. Subsequently, all mice were mated and detected vaginal plug on the second day. Mifepriston (0.8g/L 0.1ml) was administrated subcutaneously on pregnancy day 4 (10:00) to establish animal models of implantation dysfunction in Model group and TCM group. On the fifth day of pregnancy, the mean number of implanted blastocyst was compared between groups while the endometrium was dyed by Evan’s blue. The uteruses and flushings of mice in each group were obtained. The protein level of LIF in flushings were meaure by ELISA. The expression of LIF, ERa, PRa mRNA was analyzed by means of Real-time RT-PCR.Results Compared with the normal group, the coloration of endometrium dyed by Evan’s blue as well as the mean points of response was remarkably lowered in model group, and increased in TCM groups depending on dosage; The difference between normal group and high TCM group was insignificant. The level of PRa of high TCM group was higher than normal group; The level of LIF protein and mRNA of TCM group was higher than model group, furthermore, the level of LIF mRNA of high TCM group was significantly higher than normal group.Conclusions YiKang decoction can increase the pregnancy rate and implanted number of mice with embryonic implantation dysfunction.The recipe can regulate the contents of estrogen, progestogen receptors in uterus during the implantation of embryo. It can induce the expression of LIF at the right time and promote the dialogue between endometrium and embryo.The results mentioned above manifest that YiKang decoction may play an important role in the process of apposition and adhesion of the blastocyst to the uterine lumen, penetration of the epithelium and decidualization of the stromal cells.
