Abstract


In recent years, the incidence of tuberculosis infection and more serious. Bovine tuberculosis and human tuberculosis are closely related. Bovine TB can be transmitted to people, people seriously restricting the presence of tuberculosis bovine tuberculosis completely eliminated. Bovine tuberculosis can infect humans, the presence of bovine tuberculosis serious threat to human health. Therefore, strengthening the monitoring of bovine tuberculosis, bovine tuberculosis control is very important. TB diagnosis depends on pathologic examination, but required a long cycle, the positive rate is not high. Intradermal allergy diagnosis is China's legal diagnostic method, but there are many deficiencies. This paper attempts to present diagnostic methods commonly used in China and the international advanced diagnostic methods were compared in order to promote our country raise the level of bovine tuberculosis quarantine. Meanwhile, the selection bovine tuberculosis-positive cattle for M. bovis separated, and the application of advanced genotyping methods for identification. 1 Comparison of diagnostic methods abroad: Simultaneous application of EU comparative intradermal allergy, traditional intradermal allergy test for 200 cows, three days after the acquisition of the 200 head of cattle heparin whole blood, the whole blood culture, antigen stimulation with BOVIGAMTM interferon (IFN-γ) detection kit supernatant. Results 200 cows, the more positive intradermal allergy cattle, the traditional intradermal allergy-positive cattle and γ-interferon-positive cattle were 91, 98 and 95, γ-interferon with conventional allergy testing to compare allergy compliance rates were 92.86% and 95.79%. In addition, three cows in the traditional allergy test was positive in comparison with negative allergy test to prove that the traditional poultry Mycobacterium bovis allergy interference. 2 Isolation and identification of Mycobacterium bovis: Choose two domestic tuberculin, import intradermal tuberculin allergy and γ-interferon were positive cattle, autopsy, collection lungs, lymphatic lesions bacteria were isolated. After 5 days incubation, both mycobacterial growth, the growth of bacteria inoculated on Lowenstein-Jensen medium expanding culture, acid-fast staining were orchid's positive. With DNeasy Tissue Handbook kit simultaneously extracted from human tuberculosis Rv37, standard bovis strains and two clinical isolates DNA. Human tuberculosis Rv37, standard bovine strain DNA as a template, with the 16s RNA, Rv0577, IS1561, Rv1510 (RD4), Rv1970 (RD7), Rv3877 / 8 (RD1), and Rv3120 (RD12), 7 primer create Mycobacterium bacillus group stereotypes PCR method. Using the established stereotypes PCR method to extract the clinical isolates were amplified by comparison with the standard strain determined after separation of the bacteria Mycobacterium bovis were named as M.bovis-Changping07101 and M.bovis- Changping07102.
