Construction of Streptococcus suis type 2 (Streptococcus suis type 2) the virulent strain 05ZYH33 binary signal the transduction system 2148hk/rr gene knockout mutants. Build spectinomycin resistance gene in the middle, on both sides of the for 2148hk/rr coding genes downstream homologous sequences knockout plasmid screening 2148hk/rr encoding gene by homologous recombination knockout mutants. PCR analysis and Southern blot analysis showed 2148hk/rr encoding gene completely replaced by spectinomycin resistance gene knock-out in addition to successfully constructed mutants. Screening to get 05ZYH33 component signal transduction system 2148hk/rr gene knockout mutant △ 2148hk/rr laid the foundation to elucidate the regulatory system's role in the pathogenicity of Streptococcus suis. First select the different strains of mice to assess the susceptibility of 05ZYH33. 10 each of 28-day-old BALB / c and ICR strains of mice by intraperitoneal inoculation bacteria liquid per 1mL (approximately 5 × 10 8 sup> CFU). Inoculation 14 days, BALB / c mice group full death 6 ICR mice group survival, suggesting more susceptible BALB / c mice 05ZYH33, can be used for assessment of pathogen virulence.
