Effect of the Combination of Oxygen and Salvia Miltiorrhiza to Postoperative Bowel Adhesion: An Experimental Study

Abstract


Objective To observe the preventing effects of oxygen combined with salvia miltiorrhiza postoperative bowel adhesion of rabbit and explore the potential mechanisms.Methods 56 healthy male female rabbits were randomly divided into seven groups:normal control group (n=8), control group of model (n=8), positive control group (Salvia miltiorrhiza group) (n=8), gaseous group(air group(n=8),50% oxygen group(n=8)), group of gas and liquid(combining group of air and Salvia miltiorrhiza(n=8), combining group of 50% oxygen and salvia miltiorrhiza(n=8)). Except the normal control group, the bowel adhesion model was established in the rest of the groups. Normal control and model groups were feed for 8 days.Intraperitoneal injection of salvia miltiorrhiza was administered to the rabbits in the salvia miltiorrhiza’s group; intraperitoneal injection of air was administered to the rabbits in the airy group; intraperitoneal injection of 50% oxygen was administered to the rabbits in the group of 50% oxygen; intraperitoneal injection of air and salvia miltiorrhiza was administered to the rabbits in the combining group of air and Salvia miltiorrhiza; intraperitoneal injection of 50% oxygen and salvia miltiorrhiza was administered to the rabbits in the combining group of 50 % oxygen and salvia miltiorrhiza.They were all treated for 7 days and once a day.On the eighth postoperative day the blood samples of each group were collected.Grades of intestinal adhesion were ranked by macroscopic observation.The adhesive tissues between viscera and belly wall were taken for pathological observation.The levels of tumor necrosis factor-a (TNF-a) and interleukin-6(IL-6)were measured by enzyme linked immunosorbent assay.Results 1.The contents of serum TNF-a:the serum level of TNF-αin model group were significantly higher than those in normal control group(P<0.01);the contents of serum TNF-αin salvia miltiorrhiza group and treated group were significantly lower than those in model group(P< 0.01);no marked difference between the air group and the salvia miltiorrhiza(P>0.05); the contents of serum TNF-ain 50% oxygen group were significantly lower than those in salvia miltiorrhiza group (P<0.01); the contents of serum TNF-αin 50% oxygen group were significantly lower than those in air group (P<0.01); the contents of serum TNF-αin group of gas and liquid were significantly lower than those in salvia miltiorrhiza (P<0.01); the contents of serum TNF-αin combining group of50% oxygen and salvia miltiorrhiza were significantly lower than those in combining group of air and salvia miltiorrhiza (P<0.01).2. The contents of serum IL-6:the serum level of TNF-αin model group were significantly higher than those in normal control group(P<0.01);the contents of serum TNF-ain salvia miltiorrhiza group and treated group were significantly lower than those in model group(P<0.01);no marked difference between the air group and the salvia miltiorrhiza(P>0.05); the contents of serum TNF-αin 50% oxygen group were significantly lower than those in salvia miltiorrhiza group (P<0.01); the contents of serum TNF-ain 50% oxygen group were significantly lower than those in air group (P<0.01); the contents of serum TNF-αin group of gas and liquid were significantly lower than those in salvia miltiorrhiza (P<0.01); the contents of serum TNF-ain combining group of50% oxygen and salvia miltiorrhiza were significantly lower than those in combining group of air and Salvia miltiorrhiza (P<0.01).3. The situation of postoperative intestinal adhesion:the extents of postoperative intestinal adhesion in model group were significantly higher than those in normal control group(P<0.01); the extents of postoperative intestinal adhesion in salvia miltiorrhiza group and treated group were significantly lower than those in model group (P< 0.01); no marked difference between the air group and the salvia miltiorrhiza(P>0.05); no marked difference between the 50% oxygen group and the air group (P>0.05); the extents of postoperative intestinal adhesion in group of gas and liquid were significantly lower than those in Salvia miltiorrhiza group (P<0.01); no marked difference between the combining group of 50% oxygen and salvia miltiorrhiza and the combining group of air and salvia miltiorrhiza (P> 0.05).Conclusion Oxygen and the combination of oxygen and salvia miltiorrhiza all can ruduce the extent of postoperative intestinal adhesion,obviously decrease the levels of TNF-α,IL-6; the reduction was especially significant in the combination of oxygen and salvia miltiorrhiza group.
