Background: Autologous fat grafts are used for soft-tissue augmentation of various anatomic regions, most frequently for the improvement of facial contours, for their various of advantages, for example, we can get rich source of fat tissue easily, which need no more difficult operation technology, and the nice appearance and no rejection make autologous fat grafts become an ideal soft-tissue filling material. Platelet-rich plasma (PRP) is an concentration of platelets in concentrated autologous plasma, which is widely used to promote soft and hard tissue healing. The significance after using it refers to the abundance of growth factors present in a well-prepared PRP concentrate. These growth factors enhance the quality and rate of wound healing by different mechanisms. But until now, there is no report about angiogenic effect of PRP at early stage of adipose grafts.Objective: Through establishing a rabbit adipose-transplantation model, injecting combination of adipose and platelet- rich plasma (PRP) , into subcutaneous tissue on the back of nude mice. To observe angiogenic effect of platelet-rich plasma on microvessel formation at early stage after adipose graft.Method: The animals used were six adult female New Zealand white rabbits. 25ml of every rabbit’s blood was obtained from heart blood, and 20ml of it was prepared by centrifugation to make platelet-rich plasma, while 3ml of it makes the whole blood serum. The platelets were counted by platelet cell count board. Resecting the cervicodorsal adipose tissue of the rabbits was being done simultaneously. And 24 nude mices were separated into 4 groups randomly, and the fat grafts from the rabbits were injected into subcutaneous tissue on the back of the nude mices for 3 points of each one.①0.3ml pearl adipose +125mm~3gelatin sponge + 0.1ml PRP +0.1ml normal saline;②0.3ml pearl adipose + 125mm~3gelatin sponge +0.1ml whole blood serum + 0.1ml normal saline;③0.3ml pearl adipose +125mm~3gelatin sponge +0.2ml normal saline. The grafts were observed by HE and CD34 immunohistochemistry staining 1, 2, 4 and 12 weeks after operation. Microvessel density (MVD) was measured by Weidner method. All the results were statistically analyzed by the SPSS16.0 software for(x|-)±s analysis. And p﹤0.05 was considered to be statistically significant.Results: All of the injection sites were not infectious. The number of the platelet in the whole blood was (380.3±50.6)×10~9/L, while in the platelet-rich plasma was (1744.2±599.6)×10~9/L, which is 4.59 times of that in the whole blood. The MVD PRP group is significantly more than that of other groups (P﹤0.05) in the 1 and 2 week groups, but had no significant differences in the other two groups.Conclusion: PRP can significantly promote microvessel formation at early stage after adipose graft.
