Abstract


Since the oxidate cleavage of DNA activity of bis-phen copper(Ⅱ) complex was discovered, the studies on the interaction of copper complexs with DNA are attracting great interest in the field of molecular biochemistry. The studies on interaction models and mechanism of metal complexes with DNA are very important in the fields of cantineoplastic pharmaceuticals, molecular biochemistry and bioengineering technology. Recently, it is revealed that thiourea derivatives exhibit wide ranging biological activities, which including anticancer, antiviral and antibacterial, etc. Relying on the good chelate ability, thiourea and its derivatives also could be used in the extraction of trace metal and the separation of platinum group metals, and may be applied in the anion acceptor recognition. Moreover, they are wildly used in nonlinear material. Thiourea derivatives also act as common corrosion inhibitors. In this thesis, the thioureas were chosen to be ligands, and a series of new copper complexes were obtained. The crystal structures, coordination features and interactions with CT-DNA of these complexes were investigated systematically. Main contents of the thesis are given as follows:1. Review. Recently the copper complexes cleavage of DNA were systematically summarized, mainly introduces the structures and corresponding metal-nucleoenzyme activities of the copper complexes. The mechanisms of copper complexes cleavages of DNA were discussed preliminary.2. Choice and synthesis of the ligands. The basic biochemical properties and general synthesis methods of thioureas were introduced. Synthesis methods and IR characterizations of six kinds of thiourea ligands in the thesis were given in detail.3. Synthesis and spectrum characterization of copper complexes. Five copper complexes were obtained through routine methods, the single crystals were cultivated by free volatilization or diffusing way. These complexes were characterized by IR analyses.4. Analyses of crystal structures and the biochemical properties researches of these complexes. The crystal structures of three complexes were obtained by X-ray single crystal diffraction method, and the crystal characters were described. Interactions of complex (1) in crystalline A and crystalline B with CT-DNA were investigated systematically through cyclic voltammetry, UV-Vis absorption spectroscopy, fluorescence spectroscopy and viscosity measurements.
