Abstract


Rhizoma dioscoreae is one kind of herbaceous perennial vine,the stem belongs to Dioscorea opposite Thunb.It is good for spleen,lung and kindey.Polysaccharides is the main active ingredient in Rhizoma dioscoreae,which has the function of enhancing immune function,reducing blood sugar and blood fat,anti-oxidation;anti-aging and so on.This article chooses the Rhizoma dioscoreae produced in DongTai Jiang Su Province,and study the Polysaccharides in order to provide the theorical evidence to develop the source of Rhizoma dioscoreae.The article studies the extraction technology of Rhizoma dioscoreae polysaccharides. Single factor test and orthogonal experiment design method L9（34） were applied.The optimal extraction technology of Rhizoma dioscoreae pieces is with water 12 times for 3H,100℃, pH=8,polysaccharides solution concentrate to 1/5,the volume of 95%ethanol is 3 times more than polysaccharides concentrated liquid.The extraction rate of polysaccharides of Rhizoma dioscoreae is 11.58%,and polysaccharide content is 67.03%.Three methods are used to remove protein of Rhizoma dioscoreae polysaccharides.The optimal factor for removing protein is:RDCP solution 10ml（1mg/ml）,TPS 0.5ml, decomposing time 45min and decomposing temperature 37℃.The removing rate for protein is 88.7%and retaining rate for Polysaccharides is 93.1%.The Rhizoma dioscoreae polysaccharides is done by dismissing protein and pigment, flow dialysis and DEAE-52 cellulose column chromatography,the Rhizoma dioscoreae polysaccharides was separated 2 parts,RDPd-Ⅰand RDPd-Ⅱ.And then they purified by Sephadex G-100 column chromatography,RDPs-Ⅰand RDPs-Ⅱare received.They are single symmetrical peaks detected by HPLC.It concluded that the two components of Rhizoma dioscoreae polysaccharides are pure.The article studies the RDPs-Ⅰand RDPs-Ⅱby UV,IR,HPLC and GC-MS.They showed characteristic absorption peaks on UV and IR.It cannot see the 810 cm-1 and 870 cm-1 absorption peak,so it concluded that the RDPs-Ⅰand RDPs-Ⅱdo not have Man.The molecular weight of RDPs-Ⅰis 7.62×103Da,RDPs-Ⅱis 6.74×104 Da.The article studies pharmacological activity of Rhizoma dioscoreae polysaccharides.The crude Rhizoma dioscoreae polysaccharides has the function of reducing blood sugar and blood fat of diabetic mice improving glucose intolerance and liver glycogen content.RDPs-Ⅰand RDPs-Ⅱpromoted the HepG2 cell at the low level.RDPs-Ⅱhas a certain degree of inhibition of HepG2.
