Abstract


Objective:To investigate the expression of Aiolos gene and immunoglobulin IgE on asthmatic rat and the mechanisim of dexamethasone(DXM) intervention by measuring the expression of Aiolos mRNA in the peripheral blood lymphocytes and IgE in the blood of asthmatic rat and that after DXM intervention. To analyze the relationship of Aiolos gene and immunoglobulin IgE in asthma mechanisim in order to further explore the pathogenesis of asthma.Methods:Thirty SD rats were randomly divided into three groups with 10 rats in each group:the control group.the asthma group, and the DXM group. The asthma rats model were established by immunized and challenged with OVA.On the 1st day and the 8th day the asthma group were given an intraperitoneal injection of 1 ml 1% OVA saline complexed with 10% AL(OH)3. From the 15th day,ultrasonic nebulization with 1% OVA saline were used in the asthma group for thirty minutes every day,it repeated fourteen days.The DXM group were intraperitoneal injection DXM 1 hour before ultrasonic nebulization every time.The control group sensitized and stimulated with saline instead of OVA.24 hours after the last challenge, the venous blood and the lung tissue were collected under anesthesia.The establishment of asthma model was judged by the hematoxylin and Eosin (HE)stain of lung histologic section of rat and the symptoms of rat. The expression of Aiolos gene was detected by RT-PCR method. IgE level in peripheral blood was determined by an ELISA test.Results:The rat model of asthma were established successfully. Compared with the control group, the asthma group had a obviously asthma-like symptoms:shortness of breath, abdominal cramps, and cyanosis. And the bronchopulmonary histologic observations of the asthma group showed the number of eosinophils.lymphocytes, neutrophils and other inflammatory cells were increased obviously,which indicated that the rat asthma model was established successfully. Compared with the asthma group, there were less asthma-like symptoms in the DXM group. The control group had no obvious abnormalities. RT-PCR showed that the expression of Aiolos decreased in asthma group compared with control group(P<0.05), Compared with the asthma group, the expression of Aiolos in DXM group was significantly increased (P<0.05),but compared with the control group, the expression of Aiolos gene in DXM group had significant difference (P<0.05).The level of immunoglobulin IgE in peripheral blood lymphocytes of asthma rats obviously increased compared with control group (P<0.05), compared with the asthma group,the IgE level of the DXM group decreased significantly (P<0.05).however, compared with control group,it had significant difference (P<0.05).Conclusions:1. Sensitized and challenged by OVA with aluminum hydroxide.the asthma model could be established successfully.2.The expression of Aiolos gene decreased significantly in the peripheral blood lymphocytes of asthma rats.3. DXM could augmente the expression of Aiolos gene.4. There existed negative correlation between the expression of Aiolos gene and the level of immunoglobulin IgE.
