Objective:To investigate the effects of UCP2 and its posssible mechanisms in IR induced renal injury of mice by examing the expression of UCP2 mRNA and protein as well as its relationship with MDA.Methods:Twenty-eight male C57/BL6 mouses were divided randomly into normal control、IR1、IR2 and IR3 groups(n=7).All of the mouses were intraperitoneal injectied pentobarbital for anesthesia and given abdominal midline incision before the bilateral renal artery freelR groups were established by ligation of two-side pedicles for 30 minutes, and followed by 30mu、60min、120min of reperfusion respectively.Normal control group received the same operation without ischemia reperfusion.The level of UCP2 mRNA was tested by RT-PCR and protein by Western blotting.The level of MDA was tested too.Results:MDA、UCP2mRNA and protein were expressed in all the groups.IR groups received higer expression of MDA and UCP2 than the normal control group,and they increased gradually in IR groups.The results had marked difference (P<0.05).The UCP2mRNA and protein changed in parallel while MDA and UCP2 had a significant positive corelation.Conclusion:1. Ischemia reperfusion cause MDA and UCP2 expression increase.2. The expression of MDA and UCP2 increased along with the extension of time of reperfusion.3. The expression of UCP2 may be involved in the mechanism of oxidative injury in IRI.
