Abstract


Objective: To investigate the low temperature machine perfusion fluorocarbon oxygen carrying solution to save the relationship between the factors of low-flow reperfusion injury in the donor process , and provide the basis for donor cryogenic machine perfusion preservation set of traffic . Method : isolated rat liver preservation reperfusion model randomized to 0.4 ml / (min · g), 0.2ml / (min · g) and 0.1 ml / ( min · g ) flow fluorocarbon oxygen carrying liquid cryogenic machine perfusion preservation donor liver for 18 hours. Determination of low-temperature machine perfusion preservation graft reperfusion effluent alanine aminotransferase ( ALT ) , aspartate aminotransferase (AST) and endothelin (ET) content , light microscopy and electron microscopy of hepatocytes and sinusoidal endothelial cells morphological change . Separate perfusion save the group and the normal group of 0 hours . Cryogenic machine perfusion preservation the donor liver damage degree of the impact with the normal control group, these indicators to understand the factors of low flow . Results: Compared with the normal group , the control group 0.4 ml / (min · g) group , test group 0.2 ml / (min · g) and 0.1 ml / ( min · g ) group reperfusion effluent ALT, AST and ET levels were significantly higher (P <0.05), but 0.1 ml / (min · g) group increased more apparent . 0.4 ml / (min · g) group comparison , 0.2 ml / ( min · g ) group these indicators no obvious difference ( p- > 0.05 ) , 0.1 ml of / ( min · g ) group, these indicators differences significant significantly (P <0.05) . Donor tissue pathological changes of 0.4 ml / (min · g) and 0.2 ml / (min · g) group was significantly lighter than 0.1 ml / (min · g) group . Conclusion : fluorocarbon oxygen-carrying liquid cryogenic machine perfusion saved from the process of rat liver hepatocytes and sinusoidal endothelial cell damage is so severe that reperfusion when The perfusate traffic reduced to 0.1 ml / (min · g) damage serious.
