Abstract


Hepatic cancer (HC) is generally characterized by malignant cell proliferation and growth, and the occurrence of HC covered the stages of non-specific injury of liver, liver fibrosis, liver cirrhosis, dysplasia nodules and liver carcinoma. To explore the correlation between gene expression profile of rat livers and the occurrence of liver cancer at transcriptional level, a rat model of DENA-induced HC was established, and Rat Genome 230 2.0 Array was used to detect gene expression profile of liver tissues from male rats following 5, 8, 12, 16, and 18 weeks of DENA induction. To provide insights into the mechanisms of Liver cancer, Rat Genome 230 2.0 Array was used to detect gene expression profile of liver tissues from male rats following 0, 2, 6, 12, 24, 30, 36, 72, 120 and 168 hours of partial hepatectomy. Meanwhile, take JNK、ERK1/2 for example, predict the faction and difference of them in the formers by systems biology methodsThe result of Rat Genome 230 2.0 detecting gene expression profile of liver tissues from male rats following different period of DENA induction shows that 909 known genes are related with HC. and a total of 23 physiological activities, including stimulus, inflammation and immune response, signal transduction, gene transcription, metabolism of carbohydrate, lipid, amino acid, and organic acid, oxidative reduction, cell proliferation, growth, migration and adhesion etc. The results of comparing HC with LR suggested that drug/toxin metabolism and oxidation reduction were induced in HC, but decreased in liver regeneration (LR) at the transcription level, lipid, steroid and chemical homeostasis were remarkably repressed in HC but induced in LR, defense responses and apoptosis were not obvious or weaker in HC than in LR, and the activities of angiogenesis and cell adhesion/migration in HC were much stronger. Predicting the physiological activities of JNK、ERK1/2 signal pathway by the profile of gene expression indicated that JNK signal pathway increase cell apoptosis in the start-up of LR, increase cell apoptosis and proliferation in the Middle and Later Periods of LR, increase the cell proliferation in the early induction of HC, increase cell apoptosis and the cell proliferation in the middle induction Periods of HC, ERK1/2 signal pathway increase cell proliferation in the early middle of LR, increase cell proliferation in the middle induction Periods of HC.In summation, HC is closely related to 909 genes and 23 physiological activities, compared with LR, HC has widly similarities and differences, and in JNK、ERK1/2 signal pathway, the physiological activities of HC and LR have widly similarities and differences too.
