Abstract


In the past decade, Biodegradable materials that are capable of in situ formation have attracted increased attention for use in restorative orthopedic devices. For example, aliphatic polyesters, fatty acid polyester, aromatic polyester, polyamide, Polyester urea et al.In this communication, the surface erosion biodegradable polymers derived from 1.OG-polyamidoamine-double bond (PAMAM-DB) and methacrylated sebacic anhydrides (MSA).Firstly, in this paper, using methanol as solvent and ethylenediamine the initiated core, a series of dendritic Polyamidoamine(PAMAM, 0.5G-1.0G)were successfully synthesized via a divergent method,98 %. The detection results of the 1H NMR,13C NMR, FTIR confirmed the existence of various kinds of characteristic functional and terminal groups of the as-prepared PAMAM.Secondly, Methacrylated sebacic anhydride (MSA) monomer was synthesized from sebacic acid (SA) and methacrylic anhydride. The detection results of the FTIR confirmed the existence of various kinds of characteristic functional and terminal groups of the MSA.Thirdly, copolymers were synthesized form PAMAM-DB and MAS by photocrosslinking. Polymers were characterized by 1H NMR,13C NMR, FTIR, compressive strength testing and SEM. And copolymers were evaluated over 2 months period under physiological conditions. The effects of PAMAM-DB:MSA ratio on local pH, water uptake, mass loss, and mechanical properties were explored. It is found that copolymer with 50%-60% PAMAM-DB (mass fraction) show more excellent mechanical properties compared with other formulations, the whole degradation process is controlled by the surface-eroding mechanism.Lastly,1.0G-polyamidoamine-double bond (PAMAM-DB) have been blended with different amounts of methacrylated sebacic anhydrides (MSA) (50:50 and 60:40) for using them for control delivery of ofloxacin The kinetics of the drug delivery system has been systematically studied. Drug release kinetics was analyzed by plotting the cumulative release data vs. time by fitting to an exponential equation that indicated the non-Fickian type of kinetics. The drug release was investigated at different pH medium and different mass fraction of PAMAM-DB, and it was found that the drug release depends upon the pH medium as well as the mass fraction of copolymers. Our findings suggested that using PAMAM/MSA blends with different ratios, as carriers for antibiotics might be useful in the treatment of bone infection.
