Abstract

Objective: sika deer (Cervus nippon) is part of cloven-hoofed eyes, Cervidae, deer genus. The deer (Cervusnippon hortulorum) is only one of the three subspecies of northeast domestic subspecies, has high medicinal value and health value. Long been mostly concentrated in the field of physical and chemical properties, energy metabolism, feeding and management, as well as disease prevention deer, less deer nuclear gene and chromosomal studies. This article by deer R banding, restriction endonuclease significantly with its belt-type analysis and amplified fragment length polymorphism (AFLP) deer genomic DNA kinship analysis, deer cytogenetic studies to provide basic data provide some evidence of their chromosomal high-resolution G-banding and R with relationship with the AFLP and fluorescence in situ molecular hybridization (FISH) positioning and antler production traits related genes for late lay the foundation for . Method: (1) Sawing Hartshorn mining puree blood peripheral blood cells culture, synchronized cells eluted after TdR, 68 hours after adding an excess of thymidine (Thymidine, TdR) was added 5 - bromo-deoxyuridine (5 - Bromodexyuridine, BrdU) to start the cell cycle, the cells were collected, combined with RBG banding deer chromosome R-banding and analysis. (2) peripheral blood cells were cultured for 72 hours, the cells were collected, banding with restriction enzyme Hae Ⅲ and Hind Ⅲ banding chromosome specimen processing and analysis of results. (3) an ear vein blood collection, created Zabeau and Vos minor modifications amplified genomic DNA fragments of deer and amplified the results of the analysis of the genetic relationship with related software. Results and conclusions: (1) R with a single set of chromosomes banding 400. Reading Conference and livestock cytogenetic international naming system, based on the test results, drawing R with a schematic diagram of the deer. (2) two the the metacentric chromosome No1, No2, the two longest end centromere chromosome No3, No4 sex chromosomes X and the shortest, and only a sub-metacentric chromosome Y R band and high resolution G band is almost entirely the opposite, but the width have little inconsistencies. No21, No24, No28 end of the light staining, the other end are deeply stained with sambar (Cervus unicolor) the Vietnamese sika (Vietnamesesika deer) R with Y chromosome ends as darkly stained, and sheep (Ovis the Y chromosome in the R band ariesl) and goats (Capra hircus) and its end are shallow dye, which is reflected in the difference between the stability and the different species within the same species. (3) The metaphase chromosomes by restriction endonuclease Hae Ⅲ processing can show similar G-band and C-band, C band in addition to No1, No4 and 11,, No18, No27, No28, No31 several pairs of chromosomes and Y chromosomes stained shallower In addition, the shape and size of the other chromosome C with is very similar to the conventional C-band, can be in the centromere at the show deeply stained. While the the Hae Ⅲ display the G with compared with conventional G with type contrast not only weakened, but the number of bands is also relatively less, compared with conventional G-banding similar. Metaphase chromosomes by the restriction endonuclease Hind Ⅲ processing showing G-band and C-band C with addition No2, No6, 15, No18, No29, No32, several pairs of chromosomes and the X chromosome staining shallower, the rest of the chromosome morphology and The size of the conventional C band is very similar, can be in the centromere at the show deeply stained. While the the Hind Ⅲ display the G with compared with conventional G, in addition to with type contrast weakened, with its belt and conventional G are very similar. (4) 27 deer genomic DNA was amplified using AFLP technology derived fragment similarity the coefficient located between 0.6809-0.8648, with an average of 0.7841. Highest similarity coefficient between the the plucked C from Yongchuan 22 and C 20 , up to .8648 the C 22 and C 20 paternal and F 1 relationship, and paternal C 22 for high-yielding Velvet deer, their offspring F 1 20 antler production is also significantly higher than the other hybrid combinations F 1 generation higher.
