Objective: To investigate the absence of 5 - hydroxytryptamine (5-HT) deprivation of normal sleep and rapid eye movement sleep (REM sleep deprivation) Orexin - positive neuronal activity . Methods : In this study, the use of central 5-HT neurons missing conditional knock-out mice rapid eye movement sleep deprivation model mice (Pet1-Cre/Lmx1b flox / flox CKO mice) , a small platform water environmental law , immunohistochemical methods observed in wild-type mice and central 5-HT neurons observed deficient mice in state and 8 hours of normal sleep , rapid eye movement sleep deprivation after hypothalamic Orexin positive number of neurons , immunohistochemical double standard method Orexin / Fos double-labeled neurons accounted for the Orexin positive neurons proportion . Results: Orexin positive neurons before and after sleep deprivation CKO mice no significant difference compared with wild-type mice also no significant difference ; ( control group ) in the normal sleep state , CKO mice Orexin / Fos double The number of labeled neurons of wild-type mice , but significantly lower than wild-type mice sleep deprivation sleep deprivation group . Conclusion: The results of this study suggest that , as a lack of awakening to maintain the important neurotransmitter 5- HT may reduce the arousal level of the central nervous system , causing sleep deprivation can not raise the trigger and to maintain the activity of the awakening of Orexin - positive neurons .
