Abstract

Cause changes in the vaginal microenvironment during pregnancy because of the hormone levels change, decline in the number of lactobacilli, Gardnerella and some anaerobes number of increased normal vaginal flora, pregnant women prone to vaginal infections diseases, especially during pregnancy bacterial vaginosis The disease incidence increased year by year, recurrence rate, can cause a variety of adverse pregnancy outcomes. The research results show that vaginal lactobacilli maintain the pregnancy normal vaginal microenvironment plays an important role. During pregnancy bacterial vaginosis current research focuses on the application of lactic acid the Bacillus microecological treatment and screening during pregnancy bacterial vaginosis, the focus of the screening is to the vaginal PH value of screening during pregnancy bacterial vaginosis can become indicators, thus preventing adverse pregnancy outcomes occurred. In the present study, we understand the the vaginal lactobacilli content of intervention changes before and after pregnancy different gestational age through regular testing vaginal PH value, whether the application the the RAPD technology identification pregnancy vaginal flora of the Lactobacillus strains change. The first part of the quantitative detection of pregnancy vaginal lactobacilli content and RAPD Identification of Lactobacillus strains [Objective] (1) observation of changes before and after pregnancy vaginal PH value and content of vaginal lactobacilli intervention; (2) observed during pregnancy vaginal PH value and the relationship between the changes in vaginal lactobacilli content; (3) before and after the the Lactobacillus strains intervention identification pregnancy vaginal changes; [method] (1) from identifying pregnant women early pregnancy to cis production childbirth and postpartum review during timing detected vaginal PH value and content of vaginal lactobacilli; culture Lactobacillus colony count, Gram stain and random amplified polymorphic DNA analysis for the identification of vaginal lactobacilli strains; (2) pregnant women through late pregnancy period oral supplement of lactic acid bacilli after, on a regular basis to detect vaginal PH value and content of vaginal lactobacilli; Lactobacillus culture colony count, Gram stain and random amplified polymorphic DNA analysis for the identification of vaginal lactobacilli strains; Results (1) In accordance with staining showed, mainly the Lactobacillus in normal vaginal secretions large Gram-positive bacilli, arranged in single, double, short chains or palisade; (2) according to gestational different pregnancy vaginal lactobacilli colonies cultured count The results show: the late late pregnancy vaginal lactobacilli lowest content throughout pregnancy pregnancy Lactobacillus RAPD results show amplification of Lactobacillus purpose fragments in 520bp; RAPD results of the two cases better follow-up of pregnant women vaginal lactobacilli target fragment amplified Lactobacillus were 520bp at; (3) during pregnancy give oral supplement of Lactobacillus pregnancy vaginal lactobacilli colonies cultured count results show: the content of vaginal lactobacilli in 38 weeks pregnant highest, while RAPD The results show that the objective fragment amplified Lactobacillus at 520bp and vaginal lactobacilli RAPD results show, Lactobacillus amplified target fragment is still in 520bp at; preferably pregnant women in one case of follow-up at the same time. [Conclusion] pregnancy vaginal colonization by Lactobacillus strains before and after the intervention, no significant change. Second part of pregnancy vaginal PH value and vaginal lactobacilli content changes. [Objective] observed during pregnancy vaginal lactobacilli content and vaginal PH value changes between changes. 【Methods】 30 cases of pregnant women to determine early pregnancy to childbirth and postpartum review eutocia timing detection vaginal PH value, MRS agar culture colony counts statutory amount Lactobacillus content, conventional Gram stain, random amplified polymorphic DNA The analysis identified the Lactobacillus strains. [Results] (1) pregnancy pregnancy vaginal PH value change: pregnancy vaginal PH value from early pregnancy to late pregnancy period was significantly higher (PH = 3.670 VS PH = 4.005), the highest late pregnancy period (PH = 4.005 ), late pregnancy period to postpartum review during vaginal PH value decreased significantly, but still higher than during early pregnancy vaginal PH value; (2) pregnancy different pregnancy changes in vaginal lactobacilli content: pregnancy vaginal lactobacilli content decreased, which in the late stage of pregnancy to late pregnancy decreased significantly (5.042x10 ~ 7 VS 1.604x10 ~ 7), in early pregnancy to pregnancy (5.400x10 ~ 7 VS 5.042x10 ~ 7) and the late late pregnancy to postpartum two time periods (1.604x10 gently changes to 7 VS1.683x10 7); (3) during pregnancy vaginal PH value and the content of vaginal lactobacilli mutual relationship between: Gestational vaginal PH value and vaginal lactobacilli content was negatively correlated (r = -0.148, P = 0.007) . [Conclusion] pregnancy vaginal PH value increases as the pregnancy progresses, the vaginal lactobacilli content decreased as the pregnancy progresses; through the observed pregnancy vaginal PH value and content of lactobacilli change regularly detect vaginal PH value, timely exogenous supplementary lactobacilli can be effective in preventing pregnancy vaginal infections disease, reduce the incidence of adverse pregnancy. Changes after the intervention of the third part of pregnancy vaginal PH value and vaginal lactobacilli content [Objective study pregnancy and postpartum by exogenous complement Lactobacillus vaginal PH value and changes in vaginal lactobacilli content and their mutual changes relationship. 【Methods】 60 cases of pregnant women, divided into oral lactobacilli Group, the postpartum vaginal delivery group, blank control group. Oral Lactobacillus group including 30 cases of pregnant women, postpartum vaginal administration group and the control group, each group of 15 pregnant women. The oral lactobacilli group of pregnant women from 28 weeks pregnant 30 weeks pregnant first line vaginal examination after oral Lactobacillus preparation to eutocia childbirth and postpartum; the postpartum vagina administration group of pregnant women pregnant 28 weeks pregnant 30 weeks, for the first time vaginal secretions After checking during pregnancy does not give any intervention to shun production childbirth, postpartum vaginal administration given Lactobacillus preparations; blank control group of pregnant women from 28 weeks pregnant 30 weeks pregnant row for the first time after vaginal examination during pregnancy and the postpartum not give any intervention; three groups of pregnant women after taking to eutocia the timing detection childbirth and post-natal vaginal PH value and content of vaginal lactobacilli. Results (1) changes in the three groups of pregnant women during pregnancy vaginal PH value: after oral Lactobacillus group medication vaginal PH value decreased, the night before pregnancy until late late pregnancy decreased significantly (PH = 3.677 VS PH = 3.511), late pregnancy The late postpartum vaginal PH value rises (PH = 3.511 VS PH = 3.670). Throughout pregnancy vaginal PH value before taking the lowest 38 weeks pregnant. The other two groups each pregnancy vaginal PH value did not change significantly. (2) three groups of pregnant women in different pregnancy changes in vaginal lactobacilli content: the the oral Lactobacillus group vaginal lactobacilli content from the medication after to night in late pregnancy was significantly higher (4.867x10 ~ 7 VS 6.761x10 ~ 7), late late pregnancy to postpartum or (6.761x10 ~ 7 VS 6.283x10 ~ 7). The vaginal lactobacilli content throughout pregnancy at 38 weeks gestation highest minimum 32 weeks pregnant. The remaining two sets of vaginal lactobacilli content decreased with gestational age increments, each pregnancy was no significant change. (3) three groups of pregnant women vaginal the PH value Lactobacillus mutual relationship between: oral Lactobacillus group vaginal PH value and vaginal lactobacilli content was negatively correlated (r = -0.242, P = 0.008), the postpartum vaginal administration group postpartum vaginal PH value and content of vaginal lactobacilli was no significant correlation (r = 0.137, P = 0.627). [Conclusion] pregnancy oral supplement of Lactobacillus, with the advance of pregnancy, vaginal lactobacilli content increased vaginal PH value decline, helps to maintain pregnancy normal vaginal microflora.
