Abstract

Objective: 1. Lanzhou City Maternal and Child asked epidemiological survey of Candida spread. 2 to establish a rapid and simple detection and identification of Candida polymerase chain reaction (PCR) method. Discover Candida production of acid and its pathogenicity correlation, and the feasibility of the application xylitol inhibit Candida acid production and growth. Explore the feasibility of the establishment of Candida vaccine experimental animal models, experimental basis for Candida vaccine. Method: 1. Acquisition maternal vaginal secretions and oral swabs, and their newborn infants oral swabs, application CHROMagar Candida chromogenic media culture, isolation and identification of application of molecular biology techniques results verify identification. 2 the common oral candidiasis clinical samples collected by polymerase chain reaction (PCR) technology, using four pairs Candida primers amplified DNA product improvement can effectively detect common pathogens Candida PCR primers. Culture of the culture out of the four common oral candidiasis was separated application PH value detecting method detects the different time slots of each Candida acid producing force, and use the xylitol alternative medium of glucose was measured OD600 and PH . 4. Application separation cultivate a common oral candidiasis, preparation of inactivated Candida broth week fixed-dose injected into experimental mice, serum collected eight weeks after detection of Candida growth inhibition, and acquisition of the organs of the mice to observe whether the pathological damage. Results: 1. Maternal vaginal candidiasis testing positive rate was 39.42%, oral candidiasis, the positive rate was 33.65%, 21.15% of vaginal and oral positive for; neonatal oral candidiasis culture positive rate was 4.81%; 2 pairs of mother infant and simultaneous separation of Candida albicans by PCR genotype. Application of four pairs of primers a common oral candidiasis, the electrophoresis profiles based on PCR and sequencing results, confirmed by the Candida PCR primers improved than develop a high detection rate and accuracy of the method and other primers. Candida albicans, Candida tropicalis and Candida glabrata cultivate a certain time after its medium PH value is significantly lower, Candida tropicalis and Candida glabrata medium PH value after a certain period of time there is an increasing trend to continue to foster. Various Candida OD600 of the lowest in the xylitol medium; Candida albicans, Candida tropicalis and Candida krusei cultured in a medium containing glucose, pH value is significantly lower in the xylitol medium PH value no significant change. Experimental mice serum significantly inhibited the growth of Candida; observed in mice organs biopsy can also be seen, intraperitoneal injection of Candida inactivated bacterial suspension did not cause structural damage of organs and tissues of mice . Conclusion: 1. Lanzhou City neonatal candidiasis Candida detection rate of mother-to-child transmission rates higher than other regions; neonatal candidiasis infection with Candida horizontal transmission and vertical transmission. Modified multiplex PCR assay 2.ITS universal primers combined with Candida-specific primers, quick, simple, specific and sensitive detection of Candida and can be identified to species, early and rapid diagnosis of common clinical Candida treatment, has important clinical value. Candida albicans, Candida tropicalis, Candida glabrata has production of acid. Compared with other Candida, Candida albicans acid production more quickly, the acid production of the strongest and longest duration. This results the highest rate of infection of Candida clinical reports of white, the results of the most pathogenic, can be inferred pathogenic Candida certain correlation may Candida production of acid. Moreover, xylitol can inhibit Candida acid production of glucose metabolism. , Xylitol can not provide a source of nutrients for the growth of Candida, Candida growth was significantly inhibited. 4 induced experimental animal body to produce active immunity against Candida is feasible, through experimental testing, the body formed of the antibody can effectively inhibit the growth of Candida. The emergence of Candida vaccine has positive significance for the prevention and treatment of candidiasis.
