Objective : To observe the different concentrations of haloperidol on guinea pig ventricular myocytes delayed rectifier potassium current (IK) , and to explore the ionic basis haloperidol arrhythmogenic . Methods : enzymatically dissociated guinea pig ventricular myocytes , whole-cell mode voltage clamp patch clamp delayed rectifier potassium channel current role of guinea pig ventricular myocytes treated with different concentrations of haloperidol , all records are made to the same program record haloperidol intervention before and after the impact of the delayed rectifier potassium ion channel currents . Results: individual cells , were given 0.3μmol · L- 1 , the , 1.0 μ mol of L- 1 , L -1 3.0μmol haloperidol , delayed rectifier potassium ion channel currents decreased in varying degrees ; the test voltage 70mV , delayed rectifier potassium current before administration ( 16.6604 ± 1.5717 ) pA / pF , respectively, reduce to ( 5.0255 ± 1.9372 ) pA / pF ( 9.8572 ± 3.9002 ) pA / pF ( 7.1794 ± 1.816 ) pA / pF (n = 7, P lt; 0.01), and 0.3μmol · L-1 more obvious than the other two concentrations of haloperidol on the inhibition of the delayed rectifier potassium current . Conclusion: Our study found that haloperidol delayed rectifier potassium channel current inhibition , and a dose - dependent manner . Haloperidol inhibited the delayed rectifier potassium ion channel currents , action potential duration extend lead to arrhythmia haloperidol arrhythmogenic effect may be related to the role of potassium channels .
