Abstract

Objective: To establish Sprague dawley (SD) rats self-body full-thickness skin transplantation model, observations since the body of full thickness skin transplant survival process of skin grafting piece SP's content as well as epidermal stem cells in quantity, distribution changes, at the same time the naked eye observation of skin graft survival process wound of changes. Preliminary analysis of nerve fiber damage the SP content changes and skin grafting chip - the restored relations as well as the relationship of SP and epidermal stem cells and wound healing, and to explore the role of the SP in the layer of skin graft survival process. Provide a theoretical basis and experimental basis for clinical improvement in the quality of autologous skin grafts. Method: 2 to 3-month-old SD rats, male, weighing 200-250g, 63, were randomly divided into seven groups (n = 9), denoted as D0 group D1 group, D3 group, D7 group, D14 group D21 group D30 Group, D0 group for normal skin control group, the remaining six groups graft group, representing the six time limit points of autologous skin transplant recipients, that is, one day after the first 3 days, 7 days, 14 days, 21 days, 30 days. Central graft group each rat back production 1.5cm x 1.5cm size full-thickness autologous skin transplantation model, and different time points after surgery of skin grafts cut full-thickness skin tissue, normal skin control group each mouse on the back cut 1.0cm x 1.0cm size full-thickness skin tissue, without transplantation model. 1, the application of radioimmunoassay to detect the content of SP in normal skin and postoperative time point pigskin. 2, the skin tissue fixation, embedding, making biopsy, immunohistochemical method to detect epidermal stem cell surface markers β1 integrin, genetic material p63 expression observed at different time points of normal skin after skin grafting skin grafts epidermal The number of stem cells, and its distribution in the skin layers. The Block Data are presented as mean ± standard deviation (x ± s). After skin grafting group comparative study after skin grafting group carried out a comparative study between the control group with normal skin. The data were analyzed using the statistical software SPSS13.0. Results: 1, visual observation of each action?? Full thickness skin graft body the results are satisfactory, there was no obvious infection and necrosis, obvious difference in the appearance of the skin graft transplant various time limit points. 2 radioimmunoassay to detect the content of the different time points of normal skin and postoperative skin graft SP: SP content from the day after skin grafting, skin grafting chip began to decline, to three days after surgery to postoperative lowest value in a month, then the contents of SP was slowly increased until 30 days after skin graft chip SP is still lower than the preoperative level within the normal full-thickness skin. The D0 group SP content (20.88 ± 1.25) pg / mg; the D1 group SP content (10.82 ± 0.99) pg / mg; the D3 group content of SP (4.08 ± 1.18) pg / mg; the D7 group of SP content (7.50 ± 1.05) pg / mg; the D14 group content of SP (11.80 ± 1.39) pg / mg; the D21 group SP content (15.19 ± 0.80) pg / mg; the D30 group SP content (16.76 ± 1.33) pg / mg. Each group after the skin graft significant difference (P lt; 0.05), SP content in the control group of normal skin skin graft after skin grafting of SP content mean pairwise comparison except between D1 and D14 (P = 0.078 gt; 0.05), the other two groups between the number has a significant difference (P lt; 0.05). Immunohistochemical detection of pathology is divided into two indicators of β1 integrin gene substances p63. Two indicators of the rate of positive cells observed six points of time gradually decreased and reached the lowest in about three days after surgery, and then gradually increase about 7 days after the positive cell rate close to or more than the surgery reached a peak in about 21 days after surgery, and then gradually decreased. And the same time frame the same specimen serial sections of β1 integrin-positive cell rate is lower than the p63 positive cell rate of the genetic material. (1), β1 integrin: the D0 group of positive cells was (18.11 ± 1.05)%; D1 positive cells was (15.67 ± 0.87)%; D3 positive cells was (8.22 ± 1.20)%; D7 The positive cells was (17.67 ± 1.22)%; D14 group of positive cells was (19.89 ± 1.17)%; D21-positive cells was (26.44 ± 1.01)%; D30-positive cells was (20.11 ± 1.27)%. After skin grafting groups with normal skin control group, the rate of positive cells were compared, no difference in addition to the the D7 group between D0 group (P = 0.905 gt; 0.05), has among other groups with the D0 group mean significant differences (P lt; 0.05); dermoplasty?? group the average number of the positive cell rate between any two comparing D14 group and D30 between the groups (P = 0.676 GT; 0.05) there is no difference, the other both groups between the number has a significant difference (P lt; 0.05). (2), the genetic material of p63: the D0 group of positive cells was (28.22 ± 0.97)%; D1 positive cells was (25.56 ± 1.13)%; D3 positive cells was (12.89 ± 1.05)%; D7 The positive cells was (28.78 ± 1.09)%; D14-positive cells was (30.67 ± 1.32)%; D21-positive cells was (37.78 ± 1.39)%; D30-positive cells was (30.11 ± 1.45)%. Were compared in each group after skin grafting positive cell rate of the control group with normal skin, except between D7 and D0 group (P = 0.838) there was no difference, other groups and D0 were between are significant differences in resistance (P lt; 0.05); after skin grafting rate of positive cells in each group, the number of the pairwise comparison, D14 group and D30 between the groups (P = 0.336), there is no difference between the other two groups mean difference has a significant difference (P lt; 0.05). 4 positive cells in normal skin are mainly located in the basal layer of the epidermis and hair follicle bulge and flaky arrangement. Over after transplantation time, the arrangement structure of the positive cells in the skin gradually disorders, gradually distributed to the various layers of the epidermis, but D21 group began to find positive cells gradually concentrated in the basal layer of the epidermis and the hair follicle bulge in there are still positive cells in the layers of the epidermis, the D30 group was significantly less than the D21 group. Conclusion: Autologous full-thickness skin graft survival process, first to reduce the content of the SP in the skin graft piece, with the repair and regeneration of sensory nerve fibers in the skin graft piece after SP content gradually increased slowly. SP content in skin graft chip variation reflects the sensory nerve fibers in the skin graft chip damage - repair process. 2, epidermal stem cells in autologous full-thickness skin transplant survival process gradually reduce and then gradually increase, when the skin graft build enough blood supply as well as leather piece sensory innervation of gradually repair, epidermal stem cells in the epidermal cells of proportion more than normal skin. 3, the arrangement of epidermal stem cells in the skin with the progress of the process of skin graft survival showed tends to gradually disorder, almost in the epidermal layers of epidermal stem cells are able to see, after the epidermal stem cells tends to regularity within the normal skin changes, mainly in the basal layer of the epidermis and hair follicle bulge. 4, according to the variation of the content of SP and epidermal stem cells in the process of full thickness skin graft survival, combined with the naked eye changes of the wound, can be concluded that the SP and epidermal stem cells and wound healing are closely related, as well as in the process of skin graft survival SP plays an active role.
