Abstract


Objective:According to observe the postmortem degradation of high molecular-weightcaldesmon(h-CD)in stomach and appendix smooth muscle cells ofmice by Western Bloting,study the erects ofpostmortem intervals,and ambient temperature on the h-CD degradation ofmice,and the variability of the h-CD degradation in stomach smooth muscle cells and appendixsmooth muscle cells in different ambient temperature.Evaluate the application for estimatingthe time ofdeath in forensic medicine aspect.Methods:72 healthy adult Kunming mice were killed by asphyxia,and put them in 4℃refrigerator and 25℃artificial climate incubator respectively,then extract the stomach andappendix tissues at Oh,24h,48k72h,96h,120h after dead.The h-CD contents in the abovetissues were quamtitated by western-blot assay and collected by Image Pro Plus 5.0 imageanalysis system,then the datas were statistically analyzed with the SPSS 16.0 soflwaxe.Results:(1)After death,H-CD content in stomach and appendix tissues decreased with thepostmortem intervals,there is correlation between the postmortem degradation of h-CD and thepostmortem intervals;(2)The differences of the postmortem degradation of h-CD in different ambienttemperature were significant(P<0.05、.the degradation in 25℃faster than 4℃.The multipleregression equation between actin content and ambient temperature,PMl were:at 4C stomachtissues Y=14547.324-2574.543X,R2=0.997.appendix tissues Y=13326.062-2587.111X,R2=0.963;At 25℃stomach tissues Y=13917.886-2878.454X,R2=0.962.appendix tissuesY=11939.029-2663.071X,R2=0.870(Y was the IOD value of the h-CD content.X was thePMI,)(3)The differences of the postmortem degradation of h-CD between stomach andappendix tissues were significant(P<0.05),the degradation in the appendix more faster.ConelusionH-CD content in stomach and appendix tissues decreased gradually with the postmortem intervals,the postmortem degradation ofh-CD was influenced by ambient temperature,and thedegradation rate tended to be raised with higher temperature,and the differences of thepostmortem degradation of h-CD between stomach and appendix tissues were significant,thedegradation in the appendix more faster.The above analysis may be used for the estimation ofpostmortem intervals in forensic practice.
