Abstract


Objective: To investigate the effectiveness of adipose-derived stem cells(ADSCs) in prevention of expanded flap venous congestion and enhancement of the flap survival and discuss its probalble mechanisms.Methods: The stem cells were isolated from the fold inguen adipose tissue of rabbit and cultured with collagenase digestion. The passaged cells were exposed to a defined medium for osteocyte and adipocyte different- tiation. Meanwhile, tissue expander (10ml) was implanted subcutanasly in rabbits’ear. Infuse normal sodium in the expander and 1ml ADSCs (1×105/ml) were injected into the pedicle base every 7 days.One month later, dislodgment the expander and form random pattern skin flap(4cm long and 0.8cm wide).Observation the flap at 3 days and 7 days after operation.Then calculated the congestion rate and necrosis rate of the flap.Results: 1. The stem cells isolated from the rabbit’s adipose tissue could expand rapidly. The characteristics of osteocyte and adipocyte were shown after the stem cells were exposed to the defined medium.2.The congestion rate and necrosis rate in the ADSCs group was lower then that in the control group(p<0.01).Conclusion: The ADSCs of rabbits isolated in this trial are chara- cterized by stable growth and quick proliferation in vitro.Locally injection of ADSCs can prevent expanded flap from venous congestion and enhance- ment the flap survival.
