Abstract


Objective To establish OPG (osteoprotegerin, OPG) transgenic mouse model to observe the bone microarchitecture , bone mineral density and bone mineral content changes in order to investigate the OPG in vivo regulation of bone metabolism and its molecular mechanism . Constructed with human OPG gene promoter eukaryotic expression vector pCI-hOPGp-mOPG, C57BL/6J mouse zygote microinjection into prokaryotic , fallopian tubes into pseudopregnant female mice were cultured offspring mice . Mice were identified by PCR genotyping , access OPG transgenic mouse model (OPG transgenic mouse, OPG trans). Take F2 generation of SPF 10 weeks OPG trans mice and wild-type mice (C57BL/6J) of 4 , Western Blot OPG protein expression was measured in mice ; μCT mice were observed proximal tibial cancellous bone and cortical bone micro- structure ; mice was measured while applying DXA femur bone mineral density and bone mineral content in overall value . Results 1. Construct carrying molecular label Flag expression vector pCI-hOPGp-mOPG plasmid was digested with sequencing to identify the correct sequence . In 69 primary mouse , there are seven positive Founder transgenic mice . Currently F4 generation of 34 mice , there are four lines of five transgenic mice . 2 compared with wild -type mice , OPG transgenic mice tibia bone microstructure parameters of volumetric BMD , tissue mineral density , trabecular thickness and trabecular number were significantly increased, while trabecular spacing is significantly reduced ; cortical bone microstructure parameters of cortical bone area , cortical thickness , cortical bone density, cortical bone mineral content increased significantly, while the inner circumference was significantly decreased (P lt; 0.05), OD was no significant difference between the perimeter . Femur bone mineral density and bone mineral content were significantly increased (P lt; 0.05). Conclusion 1 . Successfully established molecular labels carry the OPG Flag transgenic mouse model . 2.OPG transgenic mouse cortical and cancellous bone of the bone were increased .
