Objective: Based on the establish of streptozotocin(STZ)-diabetic cataract model,this study was designed to investigate the protective effect of rosa laevigata mickx on oxidative damage in diabetic cataract rats and the antioxidant mechanism.Methods: All the lenses of 70 SD rats were examined by the slit lamp and those with any defect of lens were rejected.After 20 normal rats were taken out randomly as control,the others were injected intraperitoneally,with 1%STZ(65mg/kg) dissolved in 0.1mol/L sodium citrate buffer(PH4.5).The SD rats induced into diabetic(blood glucose>14 mmol/L) were divided into the diabetic model groups and the Rosa Laevigata Mickx treatment groups.The changes of blood glucose and body weight were monitored every 4 weeks;Progression of cataract formation in both lense of all rats was recorded using slit lamp observation every 4 weeks;At 12th weeks,the rats were sacrificed and the lenses were removed for measurement of the activities of glutathione peroxidase (GSH-PX)、catalase(CAT) and superoxide dismutase(SOD); The expression level of CuZnSOD protein and mRNA were detected by immunhistochemical method with image analyse system and RT-PCR respectively. the related comparisons and statistic analysis were carried out.Result: 1、About 80% of the rats responded to the STZ injection(blood glucose >14mmol/L).The blood glucose of diabetic model rats was statistically significant higher than that of normal control rats(P<0.05).The body weight of diabetic model rats was statistically significant lower than that of normal control rats(P<0.05);The blood glucose of Rosa Laevigata Mickx treatment groups was statistically significant lower than that of diabetic model rats(P<0.05).The body weight of Rosa Laevigata Mickx treatment groups rats was statistically significant higher than that of diabetic model rats(P<0.05).2、The transparence of the groups treated by Rosa Laevigata Mickx was higher than that of the diabetic model groups,the differences among groups were statistically significant(P<0.01).that of the normal control groups continued transparence. 3、The level of GSH-PX、CAT and SOD were decreased significantly in diabetic model groups compared with those of them in normal control groups (P<0.05),but the levels of this indexes were increased significantly in Rosa Laevigata Mickx treatment groups compared with those of them in diabetic model groups(P<0.05).4、The expression level of CuZnSOD protein and mRNA were descended significantly in diabetic model groups compared with those of them in normal control groups (P<0.05),but the expression levels of this indexes were significantly enhanced in Rosa Laevigata Mickx treatment groups compared with those of them in diabetic model groups(P<0.05).Conclusion: 1、The diabetic model induced by STZ intraperitoneal injection is reliable for the study in diabetic cataract. 2、The results showed that Rosa Laevigata Mickx can provide antioxidant effect in diabetic cataract rats.This protective effect of Rosa Laevigata Mickx may be related to the maintenance of the antioxidant status of the lens in improving antioxidant enzymes activity,mRNA and protein expression,and inhibit or delay the generation and development of cataract.
