Abstract

Objective To investigate the Han with POAG (primary open angle glaucoma, primary open-angle glaucoma) and OHT (ocular hypertension, ocular hypertension) patients FP receptor (Prostaglandin F2a Receptor prostaglandin F2α receptor, also known as FP receptor) gene polymorphism and its relationship with latanoprost drug reactions. Tier 1 screening Han POAG and OHT patients, extracted from peripheral blood specimens, and selected patients selected eye single latanoprost medication, follow-up of patients after treatment, and recorded IOP values ??and side effects. 2, using the polymerase chain reaction - restriction fragment length polymorphism (Polymerase Chain Reaction-restriction fragment length Polymorphism, PCR-RFLP) FP receptor gene rs3753380 and rs3766355 fragment polymorphism measured. 1, FP receptor in patients with POAG and OHT two single nucleotide polymorphisms (single nucleotide polymorphism) loci SNP genotype distribution the rs3753380 of C / T allele and rs3766355 of the A / C, etc. There was no statistically significant difference in the distribution of the allele (P gt; 0.05). Comparison of rs3753380 the CC CT TT genotype, the two groups in addition to the selected eye other difference was statistically significant (P = 0.006) in age, gender, diagnosis, selected eye CCT, selected visual acuity selected eye MD value was selected eye diopter and selected eye 0 days IOP differences were not statistically significant (P gt; 0.05). Rs3766355 the AA AC CC genotype, comparison, two groups in addition to the significant differences in selected eye MD values ??(P = 0.03), the rest in age, gender, diagnosis, selected eye CCT, selected visual acuity , selected eye, the selected eye refraction and selected eye 0 days IOP differences were not statistically significant (P gt; 0.05). CC AA AC genotype comparison, the two groups in addition to the selected eye CCT value differences statistically significant (P = 0.031), the rest in age, gender, diagnosis, selected eye MD values ??selected visual acuity was selected eye, the selected eye refraction and selected eye 0 days IOP differences were not statistically significant (P gt; 0.05). Medication one week, the promoter sites rs3753380 and intron sites rs3766355 genotype differences in the distribution of drug reactions without statistical significance (P gt; 0.05). Medication January the latanoprost reactive compared with other reactive difference was statistically significant (rs3753380, TT: CC CT [P = 0.014, OR = 0.80]). The conclusion first FP receptor gene promoter sites rs3753380 and intron sites rs3766355 in Han Chinese patients with POAG and OHT incidence. POAG and OHT patients of Chinese Han, rs3753380 and rs3766355 SNP distribution in Hardy-Weinberg genetic equilibrium law, still can not believe that in a small sample of the study allele distribution of two SNP loci in Han POAG and OHT patients There are differences. Rs3753380 TT genotype homozygous reactive than the non-mutated homozygous latanoprost may be higher. 4, the present study did not find a correlation between the rs3766355 SNP in Latanoprost pharmacodynamics.
