Expression and Correlation Analysis of Regulatory T Cells and NKG2D in Peripheral Blood of Esophageal Cancer Patients

Objective:Explore their expression levels and clinical significance of NK cell、activation receptor of NK cells (NKG2D) and regulatory T cells (Treg cells) in peripheral blood in patients with esophageal cancer. and in order to evaluate the dysfunction of cell immunity in patients with esophageal cancer.Method:The NK cells of mononuclear cells percentage、NKG2D expression rate and regulatory T cells of CD4+ T cells percentage in the peripheral blood, in 140 patients of esophageal cancer and 51 healthy subjects,were detected by FCM.Analyse and research on the relativity between these cells expression and the TNM classification, and the correlation of NKG2D with regulatory T cells.Results: 1.The patients with esophageal cancer, compared with the healthy subjects, had a significantly higher levels of NK (16.22±9.11%and 10.08±4.38%, respectively,P<0.01),the Nk cells expression increases with esophageal TNM clinical stage.2.The patients with esophageal cancer, compared with the healthy subjects, had significantly higher levels of Treg cells(8.39±2.04%and6.77±1.68%, respectively,P<0.001)in the peripheral blood . The expression of Treg cells rised with esophageal TNM clinical stage.3.The levels of NKG2D in the patients were significantly lower than in the healthy subjects(81.38±5.32%and85.38±3.84%,respectively,P<0.001). The expression of NKG2D decreased with esophageal TNM clinical stage .4.The expression of Treg in the peripheral blood were significantly correlated with NKG2D (r = - 0.346, P < 0.001).12% of the NKG2D total variance with Treg were concerned(R2=0.120).Conclusion:1. The NK cells of mononuclear cells percentage、NKG2D expression rate and regulatory T cells of CD4+ T cells percentage in the peripheral blood, are significantly correlated with the TNM classification in the patients with esophageal cancer.2. The regulatory T cells suppress the antitumor function of NK cells induced by NKG2D.
