Abstract


Objective:To determine the levels of regulatory T cells (CD4+T, CD4+CD25+T, CD127) and Natural killer T cells (NKT), NKG2D in patients with breast carcinoma before and after treatment, and to study their changes and clinical significance.Methods:100patients with breast carcinoma,30patients with benign hyperplasia of mammary glands, and50healthy female subjects were included this study. The levels of regulatory T cells (CD4+T, CD4+CD25+T, CD127) and NKT, NKG2D were determined in patients with breast carcinoma and benign hyperplasia of mammary glands before and after treatment, and in healthy subjects when receiving physical examination.Results:(1) The levels of CD4+CD25+T in patients with breast carcinoma before treatment were significantly increased compared with those in patients with benign hyperplasia of mammary glands and healthy subjects (P<0.01), and the levels of CD4+T, CD127, NKT and NKG2D in patients with breast carcinoma before treatment were significantly decreased compared with those in patients with benign hyperplasia of mammary glands and healthy subjects (P<0.01); the levels of CD4+T, CD4+CD25+T, CD127, NKT and NKG2D in patients with benign hyperplasia of mammary glands were not significantly different from those in healthy subjects(P>0.05).(2) The levels of CD4+T, CD127, NKT and NKG2D decreased, but the levels of CD4+CD25+T increased successively according to stage I, stage II and stage Ⅲ of tumor stage(P<0.01); the levels of CD4+T, CD127, NKT and NKG2D decreased, but the levels of CD4+CD25+T increased successively according to high differentiation, moderate differentiation and low differentiation of tumor tissue(P<0.01); the levels of CD4+T, CD127, NKT and NKG2D decreased, but the levels of CD4+CD25+T increased in patients with negative ER, positive PR, negative CerbB-2, invasive breast carcinoma and non-node-negative breast cancer were higher than those in patients with positive ER, negative PR, positive CerbB-2, non-invasive breast carcinoma and node-negative breast cancer(P<0.01or P<0.05).(3) The levels of CD4+T, CD4+CD25+T, CD127, NKT and NKG2D in patients breast carcinoma after treatment were significantly decreased compared those before treatment (P<0.01), but the levels of CD4+T, CD4+CD25+T, CD127, NKT and NKG2D in patients with benign hyperplasia of mammary glands after treatment were not significantly different compared with those before treatment(P>0.05).Conclusion:(1) The abnormal cellular immune function appears in patients with breast carcinoma;(2) The abnormal cellular immune function is closely related the occurrence and development of breast carcinoma;(3) Treatment can change the cellular immune function in patients with breast carcinoma in different level.
