Expression of HSP27 in Paclitaxel Susceptivity and Drug Resistance in Breast Cancer Cells

Abstract


Objective: Proteomic analysis of paclitaxel -sensitive and drug-resistant breast cancer cells differentially expressed proteins identified breast cancer resistant and sensitive to specific proteins reveals breast cancer chemotherapy resistant and sensitive part of the mechanism to provide a reference for clinical use chemotherapy drugs indicators provide important theoretical basis and the scientific basis for the prevention and treatment of breast cancer resistance to chemotherapy . Methods: women with breast cancer diagnosed by pathology , using the paclitaxel- cyclophosphamide program , solid tumors based on RECIST Response Evaluation Criteria in five cases each breast paclitaxel chemotherapy sensitive group and resistant group of patients screened , using two-dimensional gel electrophoresis mass spectrometry , the end of the trace protein sequencing and bioinformatics analysis of proteomics technology chemotherapy resistance group and sensitive group of breast cancer cells differentially expressed proteins , and Western blotting of HSP27 verification . Results: ( 1 ) In this study, using differential proteomics study of paclitaxel -sensitive and -resistant breast cancer cells , image analysis identified 53 differentially expressed protein spots , mass spectrometry analysis identified 40 differentially expressed protein spots identified 13 kinds of differentially expressed proteins , of which 10 species in the resistant group downregulated , three kinds of up - regulated ; ( 2 ) were detected by Western blotting of HSP27 in results proteomics results were upregulated in the sensitive group . Conclusions : (1) the establishment of 2-D DIGE map of paclitaxel -sensitive and drug-resistant breast cancer cell protein , successfully identified 13 differentially expressed proteins . These proteins for further study of the mechanisms of breast cancer chemotherapy drug to provide a theoretical basis and clues ; ( 2 ) validate HSP27 protein expression in sensitive and resistant breast cancer tissue using Western-blot technique , results and protein group analysis identified upregulated in the sensitive group . Tip HSP27 may play a role in the process of breast cancer resistance to chemotherapy , the specific mechanism of action pending further study .
