Correlation between Expression of Survivin and BCRP Gene and Effects of Neoadjuvant Chemotherapy of TE in Patients with Breast Cancer

Abstract


Objective:To study the expression changes of survivin mRNA and BCRP mRNA in breast cancer after TE regimen (Docetaxel and Epirubinciin)neoadjuvant chemotherapy and analyze the relationship between expression changes and efficiency of neoadjuvant chemotherapy,and search for biological markers to predict the effect of this neoadjuvant chemotherapy. Methods:The expression of survivin mRNA and BCRP mRA were detected by Semi-quantitative reverse-transcription polymerase chain reaction (RT-PCR) from 44 breast cancer patients before and after neoadjuvant chemotherapy of TE regimen(Docetaxel and Epirubinciin).The relationship between survivin mRNA or BCRP mRNA expression and chemotherapy response were analyzed. Results:1.The overall response rate to neoadjuvant chemotherapy was 70.5%. including 9.1%(4/44)with complete response and61.4%(27/44) with partial response. Stable disease and progression of disease were 25%(11/44)and 4.5%(2/44), respectively.2.The expression of survivin after neoadjuvant chemotherapy was 36.4%, which was significantly lower than 59.1% before neoadjuvant chemotherapy,(P<0.05).The expression of BCRP after neoadjuvant chemotherapy was 18.2%, which was significantly lower than 38.6% before neoadjuvant chemotherapy, (P<0.05).BCRP expression had significant change.3.The expression of survivin and BCRP in the effective group was lower than the negative group,there was significant differences(P<0.05).4.The expression of survivin mRNA and BCRP mRNA have no relationship(P>0.05).The efficient of the positive expression of two groups of survivin mRNA and BCRP mRNA were lower than the negative groups(P <0.05).Conclusions:TE regimen is an efficiencive neoadjuvant chemotherapy in breast cancer. The expression of survivin mRNA and BCRP mRNA influenced the effective of chemotherapy, it made the patients got poor efficacy of chemotherapy.Survivin combines with BCRP could be detected to guide chemotherapy and predict the response to it as a molecular marker in breast cancer.
