Abstract


Lung cancer is the highest cancer incidence and mortality, the low survival rate of lung cancer mainly because early diagnosis can not be effectively carried out. Sputum cytology for the early diagnosis of lung cancer, bronchoscopy and low-dose spiral CT examination, but there is over-diagnosis, lung collapse and false positive drawback. Lung cancer patients in clinical treatment, 80% are late. Highly ordered surface-enhanced Raman scattering (SERS) active substrates to improve the degree of enhancement of SERS, has been widely used in recent years due to its high sensitivity, high resolution and injury, disease diagnosis. The saliva is the ultrafiltration of the blood component and a blood considerable, has been used to diagnose a variety of diseases. This paper collected from 21 patients with lung cancer and 22 cases of normal saliva samples and measured of SERS. After spectral preprocessing, the PCA-LDA statistical analysis were obtained two samples of saliva SERS and Raman peaks analysis. PCA-LDA to obtain the accuracy rate of 86.05%, 94.12% sensitivity and 80.77% specificity. Reference tissues, body fluids and biological standard Raman peaks saliva SERS peak assigned. Changes in the two groups of samples is 523 cm -1 623 cm -1 696 cm -1 and 905 cm - 1 at, lung the patients saliva Raman peak than normal were down 27.12%, 29.58%, 33.66% and 25.74%. According to the literature, these positions Raman frequency shift may be provided by the protein in the S-S stretching vibration twist vibration and nucleic acid bases G, T, A, C ring bending vibration generated in the C-S stretching vibration, C-C. This shows that patients with lung cancer in saliva proteins and nucleic acid content on the whole is less than normal. The result The conclusions are consistent with Raman spectroscopic analysis of lung cancer cells. In the 932 cm -1 at the Raman peaks of the lung cancer patients than normal has been enhanced to more likely be attributed to the C-C stretching vibration of proline and valine literature the visible protein and nucleic acid content in the saliva of patients with lung cancer is generally lower than normal, but some of the protein content was higher than normal, or the same protein content of just a change in the conformation of certain groups. Diagnosis compared with the morphology of Medical Imaging Technology, the content of the source components within the disease and conformational changes in the protein molecules and cellular levels were detected and analyzed, with a microscopic trace, early, fast, simple, non-contact no damage, easy the Screening and dynamic monitoring of diagnostic characteristics, one direction is the development of technologies and methods of medical diagnosis.
