Abstract


Object To investigate the cause of tumor cell migration by comparing the effect of substrate stiffness on hepatic and hepatoma carcinoma cell migration so as to understand the invasive characteristics of tumor cells.Methods Microscopic morphological observation, immunofluorescence, transwell migration assays and track migration at cellular levels by time-lapse microscope were performed using human hepatoma carcinoma cell lines the HCCLM3 and human hepatic cell lines the L02.Results On 4000Pa substrate, the rates of migration and net translocation of HCCLM3 and L02 cells are higher than 500Pa and glass substrate, the mean-squared displacement of HCCLM3 and L02 cells on different substrates showed consistent tendency, and the directional persistence of L02 cells is significantly higher than HCCLM3 cells; after addition MMPs inhibitor GM6001 (40μg/mL), the rates of migration、net translocation and directional persistence of HCCLM3 cells was decreased lightly, but notably decreased in L02 cells. Intriguingly, the efficient invasion of L02 cells are lower than HCCLM3 cells in 3D environment, after addition MMPs inhibitor GM6001 (40μg/mL), the number of invasive cells was decreased in L02 cells, but notably increased in HCCLM3 cells.Conclusion In two-dimensional environment, display biphasic relationship between migration and substrate stiffness, L02 cells displayed an efficient migration due to higher directional persistence; in three-dimensional environment, the invasion efficiency of HCCLM3 cells was significantly higher due to various modes of migration adaptation to the microenvironment.
