Effect of Willed Movement Therapy on the Expression of NT-3 and GAP-43 in Penumbra Area of Rats with Cerebral Ischemia Reperfusion

Abstract


Background and Objectives intentions exercise may improve synaptic plasticity and promoting the rehabilitation of neurological function after stroke . This study in the rat model of cerebral ischemia-reperfusion injury , and with the intent to exercise therapy intervention , by observing the rat brain ischemic penumbra region NT-3 and the dynamic changes of GAP-43 to explore the intention of physical therapy on nerve regeneration and repair of its possible mechanism. Methods 240g-260g healthy and clean 54 adult male SD rats were randomly divided into model group, middle cerebral artery occlusion (MCAO group ) , raising environmental changes group (EM group ) , the intention of exercise therapy group (WM group ) , each 18 only ; using modified longa intraluminal middle cerebral artery occlusion model , in accordance with the five four points system longa neurological function evaluation reperfusion 3 days , 7 days , 15 days a total of three time points neurological deficit situation . After the rats were sacrificed by immunohistochemical staining of NT-3 protein and GAP-43 protein expression . Results 1. Neurological deficit scores : three groups of rats 3 days after reperfusion , neurological deficit scores at day 7 was no significant difference (P gt; 0.05), at 15 days after reperfusion , WM group neurological function below MCAO group and the EM group , there were significant differences (P lt; 0.05). (2) three days after reperfusion among the three groups NT-3 protein expression was no significant difference (P gt; 0.05), seven days after reperfusion and 15 days , WM group and MCAO group and the EM group were significantly different (P lt; 0.05), EM group and MCAO group showed no significant difference (P gt; 0.05). 3 reperfusion after three days , three groups GAP-43 expression was no significant difference (P gt; 0.05), seven days after reperfusion and 15 days , WM group and MCAO group and EM group were significantly different (P lt; 0.05), EM group and MCAO group showed no significant difference (P gt; 0.05). 4.NT-3 protein and GAP-43 protein expression was positively correlated . Conclusion intentions may exercise therapy by increasing the ischemic penumbra NT-3, GAP-43 expression in rat cerebral ischemia-reperfusion injury axonal regeneration , thereby promoting the rehabilitation of neurological function .
