Abstract


Objective:To observe the protective effects of edaravone to nerve cells on acute spinal cord injury, Discussing the effective treatment of second spinal cord injury with methylprednisolone for the controlMethods:Healthy SD rats of 6 months (N=144) were selected in the study. male or female, male and female rats were feed separately They were divided into two groups after acute spinal cord injury in accordance with Allen’S model by the time of the drug administration(8 hours and after 8 hours)., There are 72 rats in the first group, and then divided into 3 subgroups:A1、B1、C1, There are 24 rats in each subgroup. A1 group were given saline by the tail vein in 2、4、6、8 hours after the animal models were established as control. Six rats were randomly selected each time point; B1 group were given the methylprednisolone by the tail vein by 30mg/Kg in 2、4、6、8 hours of animal models were estabilished. Six rats were randomly selected each time point; C1 group were given the edaravone by the tail vein by 30mg/Kg in 2、4、6、8 hours of the animal models were estabilished. Six rats were randomly selected each time point; There are 72 rats in the second group, and then divided into 3 subgroups:A2、B2、C2, Every group were 24. A2 group were given saline by the tail vein in 10 12,14,24 hours of the animal models were estabilished as control, Six rats were randomly selected each time point; B2 group were given the methylprednisolone by 5.4mg/Kg/h by the tail vein in 10,12,14,24 hours of animal models were estabilished, Six rats were randomly selected each time point; C1 group were given the edaravone by the tail vein by 3mg/Kg in 10,12,14,24 hours of the animal models were estabilished, Six rats were randomly selected each time point; The two groups were both treated 2 hours. The rats of corresponding batch animals in each group were put to death. They were fixed after perfusion, then removed the injured spinal cord segment area, paraffin embedded sections after Gradient alcohol dehydration, then the Caspase-3, Bcl-xl was detected by immunohistochemistry method, three rats were randomly selected each time point,. The number of positive cells were counted. The movement function of the remaing rats which were weret reated in different time points were observed and the BBB score was recorded after the first group was treated 2 days. The movement function of the remaing rats which were were treated in different time points were observed and the BBB score was recorded after the second group was treated 2 days, using statistical software SPSS 15.0 to analysis the data of the two groups.Results:1. Motor nerve function score:Assessing the motor nerve function after the spinal cord injury by using inclined plane test.The first group(during 8 hours), the motor nerve functionof the Al group of the subgroups recovered worst,0.28±0.04, which were treated in 8th. B1 group recovered well as 0.35+0.04, which were treated in 8th.The result: B1>C1>A1, P<0.05, significant difference.The second group(after 8 hours), the motor nerve function of the A2 group of the subgroups recoved worst,0.36±0.03, which were treated in 24th. C2 group recovered well as 0.43±0.05, which were treated in 24th. The result: A2<C2, P<0.05, significant difference.2. The capase-3 positive cells increased after spinal cord injury, the first group reached the maximum in the 6th hours after the spinal cord injury. The B1 subgroup was least, the Al subgroup was most, C1 groups were between A1 and B1, B1<C<A1 (P<0.05); There were significant differences. Bcl-xl expression:C1>B1>A1(P<0.05); The positive capase-3 cells of the second group continued to increase. When the 24th hours:the C2 subgroup was least, the A2 subgroup was the most, A2>C2, (P<0.05), Bcl-xl expression:C2>B2>A2(P<0.05), There were significant differences.Conclusions:1. Edaravone has a protective effect to nerve cells on acute spinal cord and reduces secondary spinal cord injury.2.The protective effect of edaravone to nerve cells was better than methylprednisolone 8 hours after spinal cord injury.
