Abstract


Objective:By using HE staining and cell density of lumbar intervertebral disc morphology; by terminal deoxynucleotidyl transferase mediated nick end labeling (terminal deoxynucleotidyl transferase-mediated dUTP nick end labelling, TUNEL) assay lumbar disc tissue apoptosis; with streptavidin avidin - peroxidase (streptoavidin-peroxidose, SP) were detected nucleus Caspases-10 and Caspases-6 expression. Resarching the different ages of human lumbar intervertebral disc degeneration and apoptosis of the law, and analyzing the relationship between the various indicators to explore the mechanism of disc degeneration. In the future, to provide experimental evidence and treatment strategy through the possible biological method to control the lumbar disc degeneration.Materials and methods:Patient group:specimens from 45 patients were hospitalized and underwent surgery for lumbar disc herniation patients. Specimens were confirmed by pathology degenerative disc tissue. Patient group was divided into:young group (age 44 years); middle-aged group (aged 45 to 59 years); elderly group (age 60 years).Control group:5 cases of specimens were from juvenile idiopathic scoliosis orthopedic surgery patients. The specimens were taken confirmed by pathology were normal disc tissue.Patients in the surgery group were removed were L4/5 fresh nucleus pulposus; the control group specimens were removed in the surgery L2/ 3 fresh nucleus pulposus.With routine HE staining nucleus pulposus cell density and morphological changes.By TUNEL assay lumbar disc in the rate of apoptotic cells.Immunohistochemical detection of S-P method in lumbar intervertebral disc Caspase-10 and Caspase-6 expression. Antibody was replaced withPBS as a negative control.All data processing applications SPSS16.0 statistical package, test level to P<0.05 indicated significant difference.Results:1. Cell density: control group, the density of nucleus pulposus cells (8.72 ±0.66) a/HP. Middle-aged, youth, elderly patient group nucleus cell density was (7.21±0.78) a/HP; (5.79±0.60) a/HP; (4.49±0.60) a/ HP. Differences between each of four groups were significant (P<0.05). Cell density was negatively correlated with age (P<0.01).2. TUNEL positive cells:control group, TUNEL-positive cells within the nucleus was (17.80±0.62)%. Youth, middle age, elderly patients was TUNEL positive cells within the nucleus is divided into (45.71±2.05)%; (53.65±2.93)%; (68.39±4.33)%. Differences between each of four groups were significant (P<0.05). TUNEL positive cells within the nucleus and the cell density was negatively correlated (P<0.01). with age was positively correlated (P<0.01).3. The expression of Caspase-10:control group nucleus Caspase-10 positive cells was (32.60±1.64)%. Middle-aged, youth, elderly patient group nucleus Caspase-10 positive cells were (50.67±2.89)%; (63.12±4.61)%; (75.28±4.26)%. Differences between each of four groups were significant (P< 0.05).Nucleus Caspase-10 positive cells were TUNEL-positive cells rate and rate and a positive correlation between age (P<0.01). 4. The expression of Caspase-6:the control group nucleus Caspase-6 positive cells was (19.32±1.49)%. Middle-aged, youth, elderly patient group nucleus Caspase-6 positive cells was (38.58±1.80)%; (53.19±2.67)%; (62.72±4.65)%. Differences between two groups were significant in the four groups(P<0.05). Nucleus Caspase-6 positive cells were TUNEL-positive cells rate and rate and a positive correlation between age (P<0.01).Conclusion:Capase-6 and Caspase-10 protein were participated in disc tissue regulation of apoptosis. Caspase-6 and Caspase-10 may be correlated with lumbar disc degeneration.
