Abstract


Objective: experimental observations 察奈比洛尔 different artery (aorta, carotid, renal artery and femoral artery) impact on the captopril and chlorine Sha Tanshu vascular effects in spontaneously hypertensive rats (SHR). Methods: SHR homologous normotensive rats Wister-Kyoto (WKY), were randomly divided into 3 groups: (1) SHR the Nebivolol group of: nebivolol (8 mg / kg / day, ig) (2) the SHR the control group ; (3) WKY blank control group. Measurement of systolic blood pressure of the rats before the administration before the end of the experiment, at the same time. Nebivolol was dissolved in distilled water orally, the control group received an equal volume of distilled water. Measured blood pressure every week. Rat tail artery pressure measurement device (HX-Ⅱ type), non-invasive measurement of different groups of rat tail artery systolic pressure by tail cuff method in rats awake. 8 weeks after the administration, the 3% sodium pentobarbital (30mg/kg, ip) anesthetized rats, thoracotomy immediately take the aortic and carotid artery in in O2 saturation 4 ℃ PSS. Rat aortic ring, carotid ring, renal artery and femoral artery rings. Vascular tone in vitro experiments, observe the different SHR arterial vasodilator effect of captopril and losartan. Results: (1) SHR systolic blood pressure was significantly higher than their peers WKY group, after administration of nebivolol, SHR can significantly reduce systolic blood pressure. (2) arterial vasodilator captopril and losartan reaction: the SHR carotid maximal relaxation effect of captopril (E-max) and the aorta, renal artery, femoral artery losartan E-max is less than the WKY group; carotid artery of the SHR group of captopril EC50 is greater than WKY group. Nebivolol increase the SHR group of aorta, carotid artery, renal artery, femoral artery captopril and losartan, E-max, lower aorta, femoral artery captopril the EC50. Conclusion: The results showed that SHR systolic blood pressure higher than WKY and SHR carotid captopril maximal relaxant effect (Emax) is less than the WKY group (P lt; 0.05), EC50 is greater than the WKY group; the SHR group different artery ( aorta, renal artery, femoral artery) losartan maximal relaxant effect (Emax) of less than the WKY group (P lt; 0.05), but EC50 and WKY group, the difference was not significant. Nebivolol in effectively reduce the SHR systolic blood pressure at the same time, enhance the SHR arterial vasodilator effect of captopril and losartan. Increased SHR aorta, carotid, renal artery, femoral artery of captopril Emax (P lt; 0.05), reduce the aorta, femoral artery to the EC50 of captopril, carotid and renal artery EC50 was not affected. To increase the the SHR group of aortic, carotid, renal artery, femoral artery losartan Emax (P lt; 0.05), to reduce the carotid artery on the EC50 losartan aorta, renal artery and the femoral artery EC50 was not affected. To draw the following conclusions: First, the relaxation response of the SHR artery of captopril and losartan compared with WKY reduce. Second, nebivolol SHR systolic blood pressure can be significantly reduced. Three, nebivolol can increase the SHR different arterial relaxation effect of captopril and losartan.
