Abstract


Research purposes by in vitro cultured cortical cells , and low oxygen concentrations handling observe the effects of hypoxia on the cerebral cortex apoptosis ; detect cerebral cortex cells in the treatment of different low oxygen concentrations NO levels and reactive oxygen species level , explore hypoxia cerebral cortex cell apoptosis and ROS , NO to generate the level of impact mechanism . The research method within 24 hours neonatal Sprague-Dawley (SD) rat cerebral cortex was separated and in vitro cell culture to a density of 7 × 105 cells / ml , seeded in four 24 - well plates . Experimental set to 10 % , the low oxygen concentration of 0.5 % in two , each of the corresponding oxygen concentration located normoxia (20% oxygen concentration) , and each of the oxygen concentration cells inoculated with four wells , the specific packet and the hypoxic treatment as follows: cells were cultured for 24 hours after the hypoxic group corresponding concentrations hypoxia treatment , 2 hours after hypoxia , apoptosis and cellular ROS level detection by flow cytometry , ELISA assay NO . The experiment was repeated three times. Results hypoxic cerebral cortex apoptosis : hypoxia treatment group compared with normoxic control group , apoptosis percentage of 10% oxygen concentration in the treatment group was significantly greater than normoxic control group ( p lt ; 0.05 ); between different oxygen concentrations were compared , the difference does not have significant , but an oxygen concentration of 0.5% to induce apoptosis rate trend of the concentration of oxygen of less than 10% . Hypoxic cerebral cortex cells of ROS : the hypoxia treated group compared with normoxic control group , different oxygen reactive oxygen species generated by the hypoxia group were significantly lower than normoxic control group (p lt; 0.05); The comparison between the concentration difference between the non- significant . Hypoxic cerebral cortex cells of NO : hypoxic treatment group compared with the corresponding control group , NO produced by hypoxia group than normoxic control group , but the difference was non- significant presence of hypoxia -induced cell NO production trends ; compare the impact of low oxygen concentrations on cell nO , no significant difference. Research conclusions 1.10% low oxygen concentrations can increase the incidence of apoptosis in rat cerebral cortex . 2.10% , 0.5% oxygen concentration in the rat cerebral cortex cells of ROS . 3.10% oxygen concentration in the rat cerebral cortex cells to produce NO .
