Schistosomiasis is a worldwide parasite infectious diseases , has a great threat to human and livestock health and safety , the the schistosome vaccine have significant economic value and social significance . The transgenic plant vaccines is a new technology developed in recent years , because of its advantage of a cheap, safe , effective , and has become the hot spot in the field of vaccine research today . Alfalfa is the world 's most widely used forage legumes with high nutritional value and mature experience in genetic manipulation . Order to develop schistosome transgenic plant vaccines , this study will Sj23 transformed alfalfa, and DNA and the detection of the expression level of the main achievements are as follows : 1. Amplified Schistosoma japonicum Sj23 gene , inserted by TA cloning and sequencing verified pCAMBIA1303 cloning sites , constructed the eukaryotic expression vector pCAMBIA1303 - Sj23 alfalfa material LBA4404 mediated by Agrobacterium tumefaciens ( Agrobacterium tumefaciens ) , Sj23 genetic transformation of alfalfa . The use of 2 mg / L 2,4- D 0.5 mg / L KT hormone combination in MS medium induced callus , get 96% callus rate . Re-use 50 mg / L of Hyg carried out after the delay filters 120 resistant callus . 3 . Resistant callus DNA was extracted using the Sj23 primer PCR detected 68 positive calli . 10 randomly selected positive calli proteins were extracted for SDS-PAGE analysis , and found that the expression of an approximately 23kDa protein band detected by Western blot of the protein on Schistosoma japonicum infection in mice serum having immunogenicity in two calli Sj23 , indicating that may have been integrated into the genome alfalfa callus and expressed .
