Abstract


Throw yeast ( Sporidiobolus pararoseus ) JD - 2 ( Laboratory preserved strain) of the lock according to the basic principle of metabolic engineering stepwise mutagenic treatment , and ultimately to obtain a β-carotene Producing Strains JD- 2 -8 . And its solid and liquid fermentation conditions were optimized , and its application prospects . The main contents and conclusions are as follows : (1) the laboratory preserved bacteria JD-2 as a starting strain mutagenic treatment step by step through the ultraviolet (UV), microwave (MV) and sulfuric acid diethyl (DES) to diphenylamine resistance flat panel directional filtering , combined with the colony color shades final breeding high-yield strain of a β-carotene . In the non-optimized conditions under total carotenoid content increased by 0.21 mg / g dry cells to 0.57 mg / g dry cells . ( 2) lock throw yeast in a variety of carotenoids by thin-layer chromatography , liquid chromatography , LC-MS and spectral analysis method of quantitative and qualitative analysis of the final draw lock throw yeast in carotenoid composition β-carotene , the red pigment of red yeast , red yeast ene and γ- carotene , β-carotene , all-trans structure accounted for about 60% . ( 3 ) optimization of liquid fermentation conditions of β-carotene get the best medium composed of glucose 80 g / L, corn syrup 19.92 g / L , KH 2 the PO 4 < / sub > 0.96 g / L , MgSO , 4 · 7H 2 O 0.54 g / L ; optimum pH 6.0 , optimum temperature of 28 ° C . Relative dissolved oxygen control strategy for 0-16 h dissolved oxygen controlled at 20-30% ; 16-72 H 0-5% . Optimized bacteria was 56.32 g / L, total carotenoid production was 31.07 mg / L , β - carotene production up to 18.92 mg / L , 60.43% of the total carotenoids . (4) solid-state fermentation conditions to optimize the best material ratio of cornmeal : soybean meal : brewer's grains = 4:1:1 , the optimum fermentation conditions are as follows : medium optimum moisture content of 44.4% , and the optimum loading capacity for 30 g/250 mL Erlenmeyer flask , and fermentation time 108 h , the total carotenoid yield was 54.51 mg / kg dry material , β-carotene was 27.01 mg / kg dry material . Finally, analysis of solid state fermentation , the yeast carotenoids , amino acids and organic acid composition analysis of its feasibility as fodder yeast .
