Abstract


Molecularly imprinted monolithic column preparation process is simple, reproducible, column depress mass transfer speed is a very potential applications of chromatographic stationary phases. In this paper, molecular imprinting technique, caffeine, repaglinide and melamine as a template molecule using two methods of bulk polymerization and in situ polymerization synthesis of molecularly imprinted polymers and imprinted monolithic column. Effects of static balance assay to identify characteristics of the polymer and its adsorption properties. Molecularly imprinted polymer as SPE sorbent, extraction of caffeine in tea; imprinted monolithic column as the column used in liquid chromatography analysis of the repaglinide racemic chiral demolition points and milk melamine trace analysis. The paper describes the origin of the introductory molecular imprinting technology, principles, preparation and application. Coffee because the template molecule, methyl acrylic acid (MAA) as functional monomer, ethylene glycol dimethacrylate (EDMA) as a cross-linking agent, the molar ratio of the three 1 ︰ 4 ︰ 20, acetonitrile as solvent synthesized template polymer is highly selective for caffeine. Caffeine MIP made of solid phase extraction column used in the extraction of the caffeine in tea and tea caffeine content was determined by high performance liquid chromatography. The average recovery rate between 82.1% to 90.6%, the relative standard deviation is less than 5%. To Repaglinide template molecule, acrylamide (AM) as functional monomer, EDMA as crosslinker three molar ratio of 1:1 4 ︰ 20, acetonitrile as solvent, synthesis of the repaglinide highly select sexual templates polymer. Scatchard analysis showed that The repaglinide MIP repaglinide has a different affinity for the two types of sites of action, the dissociation constant of the high affinity binding sites for the dissociation constant and low affinity adsorption sites were 1.22mmol / L and 7.69mmol / L. In situ polymerization of chiral drug repaglinide imprinted monolithic column by HPLC mobile phase of acetonitrile ︰ water = 9:1 (containing 0.05% acetic acid), flow rate 0.5mL/min, detection wavelength 245nm repaglinide the MIP whole column of repaglinide and its chiral separation of enantiomers. Template molecule with melamine, MAA as functional monomer, EDMA as crosslinker three molar ratio of 1 ︰ 5 ︰ 20 toluene / dodecanol porogen, by the in situ polymerization of melamine imprinted monolithic column Determination of melamine in applied dairy. The effects of different flow relative effect of separation and optimization of chromatographic conditions selected melamine, 0.1% acetic acid solution = 5 ︰ 95 ︰ 10% methanol aqueous solution as the mobile phase, flow rate 0.5mL/min, the melamine in dairy products trace analysis. The results showed that the melamine in the range of 1.0μg/mL ~ 100.0μg/mL good linear relationship (r = 0.9987). The recovery rate was 87.0% ~ 100.5%, RSD lt; 5% (n = 6). The method limit of quantitation 1.0mg/kg, can satisfy the requirements of the Determination of Melamine in Dairy Products Limited.
