Abstract


Magnaporthe oryzae is one of the most serious ascomycete that causes rice blast, which causes great effect to the rice yield all over the world.Studying of the disease-caused basis of Magnaporthe oryzae is not only beneficial for rice blast control,but also serve as a model for revealing other fungal pathogen-plant interactions.Appressorium,the specialized cell whose structure is delicate is necessary for infection of the rice blast fungus.Apoptosis is an important form of programmed cell death,which is vital for the progress of eukaryote.The homologous of many mammalian apoptosis-related proteins have been found in yeast,and the genes and functions of these proteins have been analysised.However,until recently,the study of apoptosis-related proteins in filamentous fungi is not much.Quite recently microtubule and mitochondria interacting protein(Mmilp) has been shown to possess apoptotic functions in yeast.In this study,we indentified the hologous protein of Mmi1 in the rice blast fungus.MMI1 gene replacement vector was constructed,and hygromycin-resistance transformants were obtained by MMI1 gene fungal transformation.6 knockout mutants were identified by PCR and Southern blot. We found:Compared to Guy11,the colony modality of⊿MMI1 was much different; the growth rate and the conidium germination of mutant⊿MMI1 were suppressed;however,the pathogenicity of⊿MMI1 mutant on barley and rice was not different compared with the wild type.The⊿MMI1 mutant displays increased H2O2 resistance,which is unanimous with the results got in yeast;however,the benomyl sensitive of the⊿MMI1 mutant is identical compared with the wild type,which means that the MMI1 gene have differernt function between Magnaporthe oryzae and Saccharomyces cerevisiae.
