Expression of IL-9 in Peripheral Blood with Allergic Rhinitis Patients

Abstract


Objective:To establish a SYBR GreenⅠreal-time-fluorescence quantitative PCR method for the detection of IL-9 mRNA in human PBMC,and detect the expression levels of IL-9 in peripheral blood of allergic rhinitis (AR) patients,and analyze the expression of IL-9 in AR and observe the relationship between IL-9 and AR.Method:1) Peripheral blood mononuclear cells(PBMC) were acquired from healthy volunteers.The goal gene was amplified by PCR,and plasmid standard sample using in QRT-PCR were build by T-A clone.2) The method of SYBR GreenⅠreal-time-fluorescence quantitative PCR for the detection of this gene was established by the recombinant plasmids,which served as the standard plasmids,and the linear range, specificity and degree of precision of this method was evaluated.3) The concentration of IL-9 in plasma of AR patients was detected by enzyme linked immunosorbent assay(ELISA).4) All samples including 30 untreated AR patients,19 AR patients in clinical remission and 30 healthy volunteers were collected from the Affiliated People’s Hospital of Jiangsu University.All patients were registered gender,age.The expression levels of IL-9 were by detected the two methods.Results:1) The recombinant plasmids included 110 bps of IL-9 gene,which didn’t mutated,by sequencing.2) As for this method,the correlation coefficient of the standard curve was 0.999,the sensitivity reached 10~1 copies.The relative expression of IL-9 mRNA in the group of 30 AR patients without treatment was higher than that in the groups of 19 AR patients in clinical remission and 30 healthy controls(p＜0.05),but there were no significant differences between the groups of AR in clinical remission and normal control(p＞0.05).And the analysis of ELISA was also show the same result.Conclusions:1) SYBR GreenⅠreal-time quantitative PCR method is a sensitive method for the detection of IL-9 mRNA in human PBMC.2) The present study indicated that Th9 related cytokine factor IL-9 was higher in AR patients without treatment than healthy.That IL-9 might be involved in the pathological process of AR.The role of Th9 in process of AR remains further investigation.
