Abstract


Objective: with different concentrations of DAC intervention lung adenocarcinoma cell line A549 , to detect the survival of cells in different time observation of DAC on cell survival . 2 . Detection DAC before and after the intervention SP17 expression in lung adenocarcinoma cell line A549 , the relationship between the observed the SP17 expression and methylation , explore its expression . Detection of the SP17 lung cancer , and to explore the relationship between the expression and clinical features of lung cancer . . Study : ( 1) A549 lung adenocarcinoma cell line ( 2) to collect 40 cases of surgical lung tissue specimens , including benign lung disease , lung cancer and paraneoplastic organizations . Experimental methods : ( 1 ) MTT assay DAC intervention of cell viability before and after the A549 cells . ( 2 ) RT- PCR assay in lung tissue specimens and DAC intervention before and after SP17mRNA expression in A549 cells . (3) Western Blot detection of lung tissue specimens and DAC intervention SP17 protein expression in A549 cells before and after . Results : 1.DAC inhibit the growth of A549 cells , and drug -dependent and time -dependent . 2.DAC intervention A549 cells before and after the SP17 expression differences significantly enhanced with the decrease of the degree of methylation and expression . 3. SP17 in lung cancer and high expression in low expression in the cancer tissue , benign lung disease expression . 4. SP17 expression in lung cancer with the degree of tumor differentiation , TNM stage , lymph node metastasis-related . Conclusion : 1 . Hypomethylation status enhanced of SP17 genes and proteins expression . 2. SP17 in lung cancer, high expression associated with the pathological features of lung cancer . 3. SP17 can be used as a new biological indicator for early diagnosis of lung cancer and immunotherapy .
