Abstract


Objective:The objective of this study was to analyze the expression of Bmi-1 in hepatocellular carcinoma and its clinical significance in the prognosis of HCC patients with liver tran splantationMethods:We examined the expression of Bmi-1 mRNA and Bmi-1 mRNA and Bmi-1 protein by RT-PCR and Western blot respectively in 14 paired liver cancers and the adjacent normal tissues, which was collected from Hepatobiliary and Pancreatic surgery Departm ent of shanghai first People’s Hospital.The expression of Bmi-1 protein in 163 recipient specimens of liver cancer tissue colle ced from Pathology Department of shanghai first People’s hos pital was determined by immunohistochemistry (SABC), Statistical analyses were applied to test the relationship between expressi n of Bmi-1 and clinicopathologic features and prognosisResults:Expression of Bmi-1 mRNA and protein was detected in all cancer and adjacent normal tissue and was higher in cancers than the adjacent normal tissues. Tissue microarray immunosta ining show that the expression of Bmi-1 exist in almost every array point. high expression cases and low/-expression cases is respectively 58(35.6%),105(64.4%). statistical analysis showd expression of Bmi-1 was closely related to the tumor number (P<0.05),but not to age sex,gender,tumor size, classification, stage and AFP (P>0.05). with univariate and multivariate analysis, the expression of Bmi-1 are associated with recu rrence(P<0.001). The low Bmi-1 expression group had longer s-urvival, whereas the high Bmi-1 expression group had shorter survival. The 1、3、5-year survival rate was respectively 92.3%、98.9%,65%、86.5%, 33.9%、66.8%. Relapse-free survival rate was respectively 83.7%、93.1%, 66.7%、73.2%,21.3%、53.9%。Conclusions:This results suggest that Bmi-1 protein is expected to become not only a new biological indicators for the prognosis of HCC patients with liver transplantation,but also a targets for prevention of recurrence and therapy.
